秀珍菇子实体形成期的代谢组差异研究
Untargeted Metabolomics Profiling of Metabolic Variations During Fruiting Body Formation in Pleurotus pulmonarius
【摘要】 以秀珍菇为试材,采用基于液质联用(LC-MS,liquid chromatograph mass spectrometer)的非靶向代谢组学方法,研究秀珍菇子实体形成前后的代谢组差异,以期揭示秀珍菇子实体形成相关代谢通路和代谢标记物。结果表明:当将秀珍菇菌包低温刺激出菇直至秀珍菇原基形成整个过程分成S0、S1、S2和S3的4个不同时期,挑取4个时期的菌丝体进行基于LC-MS的代谢组分析,在正离子和负离子2种模式下共鉴定出18 262个代谢物,其中包括102个和259个、388个和269个、3 528个和4 029个差异代谢物存在于S0与S1、S0与S2和S0与S3之间。通过对代谢数据的进一步分析,表明核苷酸、氨基酸、酰胺和有机酸的代谢与秀珍菇子实体的形成密切相关,并通过多元统计分析揭示了秀珍菇子实体形成前后代谢途径和代谢物质的变化。
【Abstract】 Taking Pleurotus pulmonarius as the test material,an untargeted metabolomics study based on liquid chromatography-mass spectrometry(LC-MS)was conducted to analyze metabolic differences before and after fruiting body formation,aiming to identify key metabolic pathways and biomarkers associated with this process.The results showed that,cultivation bags of P.pulmonarius were subjected to low-temperature stimulation to induce fruiting,and the entire process-from mycelial growth to primordium formation-was divided into four stages(S0,S1,S2,and S3).Mycelia from each stage were analyzed using LC-MS.A total of 18 262 metabolites were identified in both positive and negative ion modes.Significant differences were observed in metabolite profiles,102 and 259 differentially expressed metabolites(DEMs)between S0 and S1 388 and 269 DEMs between S0 and S2,3 528 and 4 029 DEMs between S0 and S3.Further analysis revealed that nucleotide metabolism,amino acid metabolism,amide metabolism,and organic acid metabolism were closely linked to fruiting body formation.Multivariate statistical analysis highlighted substantial changes in metabolic pathways and metabolite levels during the transition from vegetative growth(S0)to fruiting body maturation(S3).These findings provided critical insights into the molecular and metabolic dynamics driving Pleurotus pulmonarius development.
