30份猕猴桃种质对碱胁迫生理响应的初步评价

【摘要】 以30份猕猴桃种质组培苗为试材,采用砂培法研究了碱胁迫对猕猴桃植株生长表型、生物量、叶片光合和根系抗氧化能力、各部位铁吸收与转运的影响,并利用主成分分析、隶属函数法和聚类分析综合评价了不同种质的耐碱性,为猕猴桃耐碱机制的解析和耐碱砧木的选育提供科学依据。结果表明,在测定的38个指标中,34个指标受碱胁迫显著影响,其中14个指标的碱胁迫敏感度大于0.6。进一步通过相关性分析筛选出9个碱胁迫响应代表性指标:株高、叶面积、根鲜重、整株干重、整株干物质含量、根全铁含量、茎与根全铁含量之比、叶与茎全铁含量之比和根POD活性。对上述9个指标进行主成分分析、基于权重的隶属函数计算和聚类分析,筛选到高耐碱型种质4份(GZ1、DE2、DE1和DE7)和高敏感型种质2份(MW3和MW13)。

【Abstract】 Thirty tissue-cultured kiwifruit germplasm vines were used to study the effects of alkaline stress on plant growth phenotype, biomass, leaf photosynthesis and root antioxidant capacity, and iron(Fe) uptake and transport in various plant parts under sand culture condition. Moreover, principal component analysis, membership function method and cluster analysis were employed to totally assess the alkaline tolerance of different kiwifruit germplasms. This will provide evidence for elucidating the mechanism of alkaline tolerance and for breeding the rootstocks in kiwifruit species. The results showed that thirty-four out of thirty-eight index tested were significantly affected by alkaline stress, and fourteen index were greater than 0.6 in alkaline sensitivity. Nine representative index were selected by correlation analysis and significant test, such as plant height, leaf area, root fresh weight, whole plant dry weight, whole plant dry matter content, total Fe concentration in roots, ratio of stem to root in total Fe, ratio of leaf to stem in total Fe, and POD activity in roots. Furthermore, the nine index were analyzed by principal component analysis, weight-based membership function method and cluster analysis, to obtain four high alkaline-tolerance germplasms(GZ1, DE2, DE1 and DE7) and two high alkaline-sensitive germplasms(MW3 and MW13).
