【摘要】 目的 探讨三七白及粉对脑出血型应激性溃疡(SU)大鼠胃黏膜的保护作用及机制。方法采用自体血定位注射法建立大鼠基底核脑出血SU模型。受试大鼠分为3组:对照组、模型组、三七白及粉组。免疫组化染色分析溃疡指数并观察胃组织病理变化。试剂盒检测大鼠血清氧化氮(NO)、丙二醛(MDA)、超氧化物歧化酶(SOD)、前列腺素E2 (PGE2)的水平。Western blot分析NLRP3抗原介导的焦亡相关蛋白(NLRP3、ASC、cleaved caspase-1和N-GSDMD)以及促炎细胞因子(IL-1 β和IL-18)的蛋白水平。结果 三七白及粉对大鼠脑出血型SU模型有良好的促进愈合作用,三七白及粉治疗可以提高脑出血型SU模型大鼠血清NO、SOD和PGE2量含并降低血清中的MDA水平。此外,三七白及粉治疗阻断了与NLRP3介导的焦亡相关蛋白(NLRP3、ASC、cleaved caspase-1和N-GSDMD)的上调以及炎症因子(IL-1 β和IL-18)的蛋白水平。结论 三七白及粉可以保护脑出血型SU大鼠胃黏膜损伤,其可能通过是阻断NRLP3介导的细胞焦亡,增强抗氧化应激作用,进而增强胃黏膜的保护作用。更多还原

【Abstract】 Objective To investigate the protective effect of panax notoginseng white powder on gastric mucosa of rats with cerebral hemorrhage stress ulcer(SU) and its possible mechanism.Methods The stress ulcer model of rat basal ganglia hemorrhage was established by autologous blood injection.The tested rats were divided into three groups:control group,model group,panax notoginseng white powder groups.HE staining was used to analyze ulcer index and observe gastric histopathologic changes.The serum levels of nitric oxide(NO),malondialdehyde(MDA),Superoxide Dismutase(SOD) and Prostaglandin E2(PGE2) were detected by the kit.Western blot analysis of NlRP3-mediated caustic death associated proteins(NLRP3,ASC,cleaved caspase-1 and N-GSDMD)and pro-inflammatory cytokines(IL-1β and IL-18) protein levels.Results Panax Notoginseng white powder and powder have a good effect on promoting healing of cerebral hemorrhage SU model.Panax Notoginseng white powder and powder can increase serum NO,SOD and PGE2 content and decrease serum MDA level in cerebral hemorrhage SU model rats.In addition,Notochiwhite and powder treatment blocked the upregulation of NLRP3-mediated pyro proteins(NLRP3,ASC,cleaved caspase-1,and N-GSDMD) and the protein levels of inflammatory factors(IL-1β and IL-18).Conclusion Panax notoginseng white and powder can protect gastric mucosa damage in rats with cerebral hemorrhage stress ulcer,which may enhance the protective effect of gastric mucosa by blocking NRLP3-mediated pyrodeath and enhancing the antioxidant stress effect.更多还原
