甘蓝型油菜胚离体胚性生长培养体系的构建

【摘要】 甘蓝型油菜种子含油量受母体基因型、胚基因型、细胞质基因型等遗传效应及大田环境因素的影响。独立研究含油量影响因素就需要建立一个维持胚性生长的胚离体培养体系。结果显示,以甘蓝型油菜ZY 511第22 DAP(Days after pollination)幼胚为材料,B 5为基本培养基,8%蔗糖、0.6%琼脂、0.05 mg/L NAA(萘乙酸)、0.15 mg/L 6-BA(6-苄氨基腺嘌呤)时,胚性生长率最高(94.64%)。以此建立的胚离体培养最佳胚性生长体系为油菜种子含油量的遗传机制研究提供了技术支撑。

【Abstract】 The oil content of Brassica napus L. seed was influenced by genetic effects of maternal genotype, embryonic genotype, cytoplasmic genotype and field environment factors. For independent research, it was necessary to establish an embryo culture in vitro system to maintain embryonic growth. The results showed that the young embryos of Brassica napus L. ZY 511 at 22 th DAP(days after pollination) were as materials, B 5 as base medium, 8% sucrose, 0.6% AGAR, 0.05 mg/L NAA(naphthoacetic acid), 0.15 mg/L 6-BA(6-benzylamino-adenine), reaching the highest embryonic growth rate(94.64%). The optimal embryonic growth system of embryo culture in vitro provided technical support for the study of genetic mechanism of oil content in Brassica napus L. seeds.
