黄花苜蓿AP2/ERF家族MfERF028基因的表达特性与耐寒功能分析

【摘要】 基于黄花苜蓿(Medicago falcata L.)转录组测序数据,克隆得到MfERF028基因,蛋白质编码区(Coding sequence,CDS)长度为672 bp,编码223个氨基酸。该基因编码的蛋白质相对分子质量为25.3 kDa,等电点为6.00;亚细胞定位表明该基因所编码的蛋白在细胞核内特异表达,基因表达模式分析表明对-8℃胁迫响应强烈。构建植物表达载体,并转入截形苜蓿(Medicago truncatula Jemalong A17)中进行基因功能验证,结果表明异源表达的转化植株遭受冷冻胁迫时MfERF028可上调MtCAS15A基因的表达。该研究为阐明AP2/ERF家族基因抗逆机制提供了理论依据,并为培育抗逆豆科牧草提供了新的候选基因。

【Abstract】 In this study, MfERF028 gene was cloned based on the RNA sequencing data of Medicago falcata L., and the coding sequence(CDS) was 672 bp in length, encoding 223 amino acid. The relative molecular weight of the protein encoded by this gene was 25. 3 kDa, with an isoelectric point of 6. 00. Subcellular localization indicated that the protein encoded by MfERF028 gene was specifically expressed in the nucleus, and gene expression pattern analysis indicated a strong response to-8 ℃ stress. The plant expression vector was constructed and transferred into Medicago truncatula Jemalong A17 for gene functional verification. The results showed that MfERF028 could up regulate the expression of MtCAS15A gene when the heterologous transformed plants were subjected to freezing stress. This study would provide a theoretical basis for elucidating the stress resistance mechanism of AP2/ERF family genes and could provide new candidate genes for cultivating stress resistant leguminous forages.
