利用DNA条形码及野生扁穗雀麦居群改进遗传学综合实验——以云南农业大学生物技术专业遗传学综合实验为例
Exploration of improving genetic comprehensive experiment by using DNA barcoding and wild Brome catharticus Vahl population——taking the comprehensive genetics experiment of biotechnology specialty in Yunnan Agricultural University as an example
【摘要】 文章对“学生采集云南农业大学野生扁穗雀麦居群数量性状数据并分析,分别利用核DNA及叶绿体DNA通用引物ITS及rbcL扩增所采集个体总DNA,电泳检测”作为遗传学综合实验教学案例的实施过程、效果及可行性进行了验证和探讨。实验以昆明常见野生扁穗雀麦作为实验材料,一个实验班的学生每人采集一株植株的株高、分蘖、有效穗数、平均穗长、平均小穗数5个数量性状,全班数据共享,进行方差分析;每个同学分别提取所采植株总DNA,一半学生利用引物rbcL、另一半学生利用引物ITS进行PCR扩增,电泳检测扩增产物。结果显示,ITS扩增片段约750bp,rbcL扩增片段约500bp。此综合性实验是充分利用地方资源,并将科研成果应用于本科实验教学的一个有益探索。
【Abstract】 The article validates the implementation, effectiveness and feasibility of "students collected and analysed quantitative trait data from wild Brome population populations at Yunnan Agricultural University, and amplified the total DNA of the collected individuals using the common primers ITS and rbcL for nuclear DNA and chloroplast DNA, respectively, and detected them by electrophoresis" as a case study of a comprehensive genetics experiment. The experiment was conducted using wild B. population commonly growing in Yunnan Agricultural University and its surroundings as the experimental material. Each student in a class collected one plant for five quantitative traits: plant height, tillers, effective spike number, average spike length and average spikelet number.The amplification results were examined by electrophoresis. 750bp and 500bp fragment were amplified by the ITS and rbcL respectively. This comprehensive experiment is also a useful exploration of the use of local resources and the application of research results to undergraduate laboratory teaching.
