1株益生乳酸菌的分离及其制备的发酵饲料对仔猪生长性能、血清抗氧化与免疫相关指标的影响

【摘要】 为了寻找新的可用于制备仔猪发酵饲料的益生乳酸菌,试验采用MRS培养基从健康仔猪粪便中分离纯化乳酸菌,利用16S rDNA基因测序结合生理生化检测对所得菌株进行鉴定。在此基础上,将分离得到的益生乳酸菌与蜡样芽孢杆菌(Bacillus cereus)PBC-1制成复合菌剂添加到基础日粮中制备发酵饲料。将120头体重相近的28日龄健康三元杂交断奶仔猪随机分为4组,分别为对照组及2%、5%、10%发酵饲料组,每组3个重复,每个重复10头。对照组饲喂基础日粮,2%、5%、10%发酵饲料组分别在基础日粮中添加2%、5%、10%的发酵饲料替代相应比例的基础日粮,预试期2 d,正试期28 d。试验结束后统计仔猪的平均日采食量、平均日增重及腹泻率,每头仔猪采血并分离血清,用相应试剂盒检测血清抗氧化指标、免疫指标及炎症因子。结果表明:从健康仔猪粪便中筛选得到1株干酪乳酪杆菌(Lacticaseibacillus casei),命名为ZYSJ-2。与对照组相比,2%、5%和10%发酵饲料组仔猪平均日采食量差异不显著(P>0.05),但平均日增重均显著提高(P<0.05),仔猪腹泻率均显著下降(P<0.05)。仔猪的血清总抗氧化能力(T-AOC)及谷胱甘肽过氧化物酶(GSH-Px)、总超氧化物歧化酶(T-SOD)、过氧化氢酶(CAT)活性均显著提高(P<0.05),血清丙二醛(MDA)浓度则显著降低(P<0.05)。2%、5%和10%发酵饲料组仔猪的血清免疫球蛋白A(IgA)、免疫球蛋白M(IgM)、免疫球蛋白G(IgG)质量浓度均显著高于对照组(P<0.05),而炎症因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)及肿瘤坏死因子-α(TNF-α)质量浓度则显著低于对照组(P<0.05)。说明筛选得到的ZYSJ-2能够用于制备猪发酵饲料,采用干酪乳酪杆菌ZYSJ-2与蜡样芽孢杆菌PBC-1制得的复合发酵饲料能够提高仔猪的生长性能,增强仔猪抗氧化能力与免疫力,提升仔猪健康水平,复合发酵饲料以5%比例与仔猪的基础日粮混合使用为宜。

【Abstract】 In order to find new probiotic lactic acid bacteria which can be used to product fermented feed for piglets, lactic acid bacteria were isolated and purified from the feces of healthy piglets using MRS medium. The strain was identified through 16S rDNA sequencing combined with physiological and biochemical experiments. On this basis, a composite inoculum of probiotic lactic acid bacteria and Bacillus cereus PBC-1 added to the base diet and was used to produce fermented feed. 128 healthy twenty-eight-day-old crossbred piglets were randomly divided into 4 groups: control group and 2%, 5% and 10% fermented feed groups with 3 replicates per group and 10 piglets per replicate. The piglets in the control group were fed the basal diet, and the piglets in 2%, 5% and 10% fermented feed groups were fed the trial diet supplemented with 2%, 5% and 10% fermented feed instead of the corresponding proportion of the basal diet, respectively. The pre-test period was 2 days and the experimental period was 28 days. At the end of the experimental period, the average daily feed intake, average daily weight gain and diarrhea rate of piglets were counted. Blood samples was collected from each piglet and serum was separated. Serum antioxidant indexes, immune indexes and inflammatory factors were detected by kit. The results showed that a strain of Lacticaseibacillus casei was isolated from the feces of healthy piglets and named ZYSJ-2. Compared with the control group, the average daily feed intake of piglets in 2%, 5% and 10% fermented feed groups was not significantly different(P>0.05), but the average daily weight gain was significantly increased(P<0.05), the diarrhea rate of piglets was significantly decreased(P<0.05). Serum total antioxidant capacity(T-AOC) and activities of glutathione peroxidase(GSH-Px), total superoxide dismutase(T-SOD) and catalase(CAT) were significantly increased(P<0.05), while serum malondialdehyde(MDA) concentration was significantly decreased(P<0.05). The mass concentrations of serum immunoglobulin A(IgA), immunoglobulin M(IgM) and immunoglobulin G(IgG) of piglets in 2%, 5% and 10% fermented feed groups were significantly higher than those in the control group(P<0.05), and the mass concentrations of inflammatory factors interleukin-1β(IL-1β), interleukin-6(IL-6), interleukin-10(IL-10) and tumor necrosis factor-α(TNF-α) were significantly lower than those in the control group(P<0.05). These results indicated that the screened ZYSJ-2 could be used for the production of fermented feed for piglets. The composite fermented feed prepared with Lacticaseibacillus casei ZYSJ-2 and Bacillus cereus PBC-1 could improve the growth performance of piglets, enhance their antioxidant capacity and immune abilities, and improve their health level. It was appropriate to mix the composite fermented feed with the basal diet of piglets at a ratio of 5%.
