一株引起广西桑根腐病的病原真菌分离及初步鉴定

【摘要】 采用组织分离法,从广西环江蚕区桑园发生根部腐烂的桑树根部分离到一株真菌菌株,命名为QYE07。根据菌丝形态和rDNA-ITS序列分析对该菌株进行鉴定,并采用浸根法将菌株接种到健康实生桑苗根部,测定其对桑树的致病性。结果表明,该菌株在PDA培养基上的菌落形状、孢子形态学特征与镰刀菌一致;经rDNA-sITS测序获得长度为532 bp的序列片段,其与镰状镰刀菌(Fusarium falciforme)的亲缘关系最近,一致性达99.8%以上;病原菌致病试验显示该菌株可引起桑树根部腐烂以及桑叶黄化、枯萎等症状,初步鉴定其为镰状镰刀菌(Fusarium falciforme)。文中关于镰状镰刀菌浸染桑树致根腐病的报道可为桑树根腐病防治提供参考。

【Abstract】 A pathogenic fungal strain named QYE07 was isolated by tissue separation method from the fibrous roots of mulberry trees with root rot onset in Huanjiang, Guangxi. This fungal strain was identified based on mycelial morphology and rDNA-ITS sequence analysis. And to test the pathogenicity to mulberry trees, pathogen was inoculated to healthy mulberry trees by root dipping method. The results showed that the appearance and spore morphological characters of this fungal strain cultured on PDA medium were similar to those of Fusarium, and a sequence fragment of 532 bp in length was obtained by rDNA-ITS sequencing. It had the closest genetic relationship with Fusarium falciforme, with a identity of over 99.8%. The pathogenic test demonstrated that this fungal strain could cause root decay, leaf yellowing and wilting in mulberry trees. Therefore, it was identified as F. falciforme. The report in this study about F. falciforme causing root rot of mulberry may provide a theoretical basis for the control of root rot of mulberry.
