miR-19a-3p联合PTEN基因在胃癌中的研究价值

【摘要】 目的 研究微小RNA miR-19a-3p及PTEN基因与胃癌的相关性,以及miR-19a-3p在胃癌组织及相应正常胃组织中的表达情况,进一步探索miR-19a-3p联合PTEN基因能否作为诊断胃癌的生物学标记物。方法 24例胃癌患者,留取术后胃癌及正常胃组织标本制备蜡块,采用实时荧光定量聚合酶链式反应(PCR)法对胃癌组织及正常胃组织中miR-19a-3p的表达情况进行检测,采用免疫组化染色法判断PTEN基因染色情况。结果 miR-19a-3p在胃癌组织中表达量高于正常胃组织。肿瘤最大径>50 mm、年龄≥66岁、男性、淋巴结有转移均不是影响miR-19a-3p高表达的单因素,差异无统计学意义(P>0.05)。miR-19a-3p在胃癌组织中高表达,而胃癌分化越差, PTEN免疫组化染色越呈阴性。结论 miR-19a-3p在胃癌组织中高表达与低分化胃癌PTEN免疫组化染色呈阴性有关,未得到miR-19a-3p可能作为胃癌诊断的生物学标记物的可靠证据。

【Abstract】 Objective To study the correlation between microRNA miR-19a-3p and PTEN gene and gastric cancer, as well as the expression of miR-19a-3p in gastric cancer tissues and corresponding normal gastric tissues, and further explore whether miR-19a-3p combined with PTEN gene can be used as biological markers for the diagnosis of gastric cancer. Methods In 24 patients with gastric cancer, postoperative gastric cancer and normal gastric tissue specimens were collected and wax blocks were prepared. The expression of miR-19a-3p in gastric cancer tissues and normal gastric tissues was detected by real-time fluorescence quantitative polymerase chain reaction(PCR), and PTEN gene staining was determined by immunohistochemical staining. Results The expression level of miR-19a-3p in gastric cancer tissue was higher than that in normal gastric tissue. Tumor maximum diameter >50 mm, age ≥66 years, male gender, and lymph node metastasis were not single factors affecting the high expression of miR-19a-3p, and the difference was not statistically significant(P>0.05). miR-19a-3p was highly expressed in gastric cancer tissues, and the poorer the gastric cancer differentiation was, the more negative the PTEN immunohistochemical staining was. Conclusion The high expression of miR-19a-3p in gastric cancer tissue is correlated with negative PTEN immunohistochemical staining in poorly differentiated gastric cancer, and there is no reliable evidence that miR-19a-3p might be used as a biomarker for gastric cancer diagnosis.
