miR-495-3p靶向IL-6R抑制宫颈癌细胞的增殖和转移的机制研究
miR-495-3p inhibits proliferation and metastasis of cervical cancer cells via targeting IL-6R
【摘要】 目的:探讨miR-495-3p和白细胞介素6受体基因(IL-6R)在宫颈癌细胞增殖和转移中的作用。方法:实时荧光定量PCR(RT-qPCR)和Western blot检测人宫颈永生化细胞H8和宫颈癌细胞(SiHa、HeLa、caski)中miR-495-3p和IL-6R表达水平。将SiHa细胞分为miR-NC、miR-495-3p、si-NC、si-IL-6R、miR-495-3p+pcDNA-NC、miR-495-3p+pcDNA-IL-6R组。细胞计数试剂盒(CCK-8)、Transwell实验分别检测SiHa细胞的增殖和转移能力。Western blot检测细胞周期素D1(CyclinD1)、基质金属蛋白酶2(MMP2)、MMP9蛋白表达。双荧光素酶报告实验、Western blot确定miR-495-3p对IL-6R的靶向调控作用。结果:与H8细胞比较,宫颈癌细胞中miR-495-3p表达降低,IL-6R表达升高(P<0.05)。过表达miR-495-3p或抑制IL-6R表达后SiHa细胞CyclinD1、MMP2和MMP9表达降低,细胞增殖和转移能力降低(P<0.05);过表达IL-6R可逆转miR-495-3p对SiHa细胞增殖、转移以及其相关蛋白表达的影响(P<0.05)。IL-6R是miR-495-3p的靶基因,miR-495-3p负调控IL-6R表达。结论:miR-495-3p通过靶向IL-6R抑制宫颈癌细胞的增殖和转移,提示miR-495-3p在宫颈癌中具有抑癌作用。
【Abstract】 Objective:To investigate the role of miR-495-3 p and interleukin 6 receptor gene(IL-6 R)in the proliferation and metastasis of cervical cancer cells.Methods:The expression levels of miR-495-3 p and IL-6 R in human cervical immortalized cells H8 and cervical cancer cells(SiHa,HeLa,caski)were detected by real-time fluorescent quantitative PCR(RT-qPCR)and Western blot.SiHa cells were divided into miR-NC,miR-495-3 p,si-NC,si-IL-6 R,miR-495-3 p+pcDNA-NC,miR-495-3 p+pcDNA-IL-6 R group.Cell counting kit(CCK-8)and Transwell test were used to detect the proliferation and metastasis ability of SiHa cells. Western blot was used to detect cyclin D1(CyclinD1),matrix metalloproteinase 2(MMP2),and MMP9 protein expression. The dual luciferase report experiment and Western blot confirmed the targeted regulation of miR-495-3 p on IL-6 R.Results:Compared with H8 cells,the expression of miR-495-3 p decreased and the expression of IL-6 R increased in cervical cancer cells(P<0.05). After overexpression of miR-495-3 p or inhibition of IL-6 R expression,the expressions of CyclinD1,MMP2 and MMP9 in SiHa cells were reduced,and cell proliferation,metastasis were reduced(P<0.05). Overexpression of IL-6 R could reverse the effect of miR-495-3 p on proliferation,metastasis and its related protein expression to SiHa cells(P<0.05). IL-6 R was validated to be the target gene of miR-495-3 p,miR-495-3 p negatively regulated IL-6 R expression.Conclusion:miR-495-3 p inhibits the proliferation and metastasis of cervical cancer cells by targeting IL-6 R,suggesting the repressive effect of miR-495-3 p in cervical cancer.
