浸种与层积过程中紫椴种子内源激素变化

【摘要】 探究紫椴种子内源激素在浸种和低温层积过程中的含量变化以及在种子不同部位的分布.利用高效液相色谱串联质谱法分析吲哚乙酸(IAA)、脱落酸(ABA)、赤霉素(GA3)、玉米素(ZT)含量变化,探讨其在打破种子休眠过程中的作用.定量分析结果表明:在浸种过程中,IAA含量一直处于动态变化,果皮中的IAA含量低于种子内部胚中的IAA含量;ABA含量呈先降低后升高的趋势,浸种8 d达到最低;浸种初期,种子中GA3含量总体较低,随着浸种时间的增加呈增加趋势;种子外部的ZT含量始终低于种子胚乳中的ZT含量,浸种2 d含量最高,4~10 d呈现先增加后降低的趋势;ABA/IAA、ABA/ZT、ABA/GA3大致呈先降低后升高的趋势.在层积过程中,种子中IAA含量呈“升高—降低—升高”的趋势;ABA含量大致呈“升高—降低”的趋势;GA3含量在层积初期呈上升趋势,于45 d左右达到最大,继续层积含量逐渐下降;种子内部的ZT含量一直高于果皮中的ZT含量,整体变化趋势呈“W”状,层积45 d达到峰值,60 d急剧下降;ABA/IAA、ABA/ZT、ABA/GA3呈先上升后降低的趋势.

【Abstract】 In order to explore the content changes of endogenous hormones in Tilia amurensis seeds during seed soaking and low temperature stratification and their distribution in different parts of seeds, the content changes of indoleacetic acid(IAA),abscisic acid(ABA),gibberellin(GA3) and Zeatin(ZT) were analyzed by high performance liquid chromatography tandem mass spectrometry.This paper discussed the function of the seed dormancy breaking.The quantitative analysis results indicated that the content of IAA in the pericarp was lower than that in the inner embryo of seed.The content of ABA decreased at first and then increased, and reached the lowest level at 8 d.The content of GA3 was low in the initial stage of seed soaking, and increased with the increase of seed soaking time.During the whole soaking process, the ZT content outside the seeds was always lower than that in the endosperm.The ZT content was the highest at 2 d after soaking, and increased first and then decreased at 4~10 d.The results showed that the values of ABA/IAA,ABA/ZT and ABA/GA3 decreased first and then increased.In the process of stratification, the content of IAA in seeds increased, decreased and increased.The content of ABA increased and decreased; The content of GA3 increased at the beginning of stratification and reached the maximum at 45 d.The content of GA3 decreased gradually after stratification.The content of ZT in seeds was always higher than that in pericarp, and the overall change trend was W-shaped.It reached the peak after 45 d of stratification, and then decreased sharply after 60 d with the increase of stratification time.The values of ABA/IAA,ABA/ZT and ABA/GA3 increased first and then decreased in the process of stratification.
