不同剂量维生素D2对膝关节骨性关节炎大鼠IL-1β和MMP-13表达的影响
Effects of different doses of Vitamin D2 on the expression of IL-1β and MMP-13 in KOA rats
【摘要】 目的 观察使用不同剂量维生素D2干预后膝关节骨性关节炎(knee osteoarthritis, KOA)大鼠关节液中IL-1β的浓度和关节软骨组织中MMP-13的表达情况,探讨其对炎症反应及软骨降解的影响。方法 选择2~3月龄的雌性SD大鼠50只,采用关节腔内注射木瓜蛋白酶法制备KOA大鼠模型,随机将大鼠分成A组(模型组)、B组(维生素D2剂量为4×105 U/kg)、C组(维生素D2剂量为5×105 U/kg)、D组(维生素D2剂量为6×105 U/kg)和E组(维生素D2剂量为7×105 U/kg),每组10只。除A组外,其余四组均给予维生素D2干预,1次/周,连续4周。4周后取大鼠膝关节组织制备切片,进行HE染色、甲苯胺蓝染色、改良的番红O-固绿染色和Mankin’s评分,检测大鼠关节液中IL-1β浓度和软骨组织中MMP-13的表达情况。结果 经过不同剂量维生素D2干预后,各组均可见软骨细胞增多,基质染色加深,软骨层增厚,Mankin’s评分、关节液中IL-1β的浓度和软骨组织中MMP 13的表达水平均较模型组降低(P<0.05)。其中,维生素D2剂量为6×105 U/kg组的软骨层增厚较其他维生素D2剂量组明显,Mankin’s评分、关节液中IL-1β浓度及软骨组织中MMP-13的表达水平均低于其他维生素D2剂量组。结论 维生素D2可通过抑制IL-1β和MMP-13的表达水平,减轻关节内的炎症反应、减少软骨基质降解,促进关节软骨修复。且剂量为6×105 U/kg的维生素D2干预效果较好。
【Abstract】 Objective To observe the concentration of IL-1β in synovial fluid and the expression of MMP-13 in articular cartilage tissue of KOA rats after intervention with different doses of Vitamin D2, and to explore their effects on inflammatory response and cartilage degradation. Methods Fifty female SD rats aged 2-3 months were selected, and the KOA rat model was prepared by intra-articular injection of papain. The rats were randomly divided into group A(model group) and group B(4 × 10~5 U/kg), group C(5 × 10~5 U/kg), group D(6 × 10~5 U/kg) and group E(7 × 10~5 U/kg), 10 in each group. Except for group A, the other four groups were given Vitamin D2 intervention once a week for 4 consecutive weeks. The rat knee joint tissue was taken to prepare sections, HE staining, toluidine blue staining, modified safranin O-fast green staining and Mankin’s score were performed to detect the concentration of IL-1β in synovial fluid and the expression of MMP-13 in cartilage tissue of rats Happening. Results After the intervention of different doses of Vitamin D2, chondrocytes increased, matrix staining was deepened, cartilage layer thickened, Mankin’s score, IL-1β concentration in synovial fluid and MMP 13 expression level in cartilage tissue were all lower than those in the model group( P<0.05). Among them, the cartilage layer thickening in the Vitamin D2 dose group of 6 × 10~5 U/kg was significantly thicker than that in other Vitamin D2 dose groups, and the Mankin’s score, the concentration of IL-1β in synovial fluid and the expression level of MMP-13 in cartilage tissue were lower than those in the Vitamin D2 dose group. Conclusion Vitamin D2 can reduce the inflammatory response in joints, reduce cartilage matrix degradation, and promote articular cartilage repair by inhibiting the expression levels of IL-1β and MMP-13. And the intervention effect of Vitamin D2 at a dose of 6 × 10~5 U/kg was better.
