利用荧光SSR标记构建欧李种质分子身份证
Establishment of Molecular Identity for Cerasus humilis Germpalsms Using Fluorescently Labeled SSR
【摘要】 以19个欧李优异种质及其近缘种为试验材料,利用荧光SSR标记构建分子身份证。从欧李近缘属植物中筛选出多态性高、稳定性好的13对引物,对19个欧李品种及其近缘种杏李、李资源的遗传多样性和亲缘关系进行分析。结果表明:共检测到等位基因数88个,每对引物检测到的等位基因数在2~12个,平均6.7个;共检测到等位基因片段98个,每对等位基因检测到的等位基因片段数在3~12个,平均7.5个。多态信息含量变幅在0.296 1~0.853 3,平均为0.629 7;Shannon信息指数变幅在0.547 4~2.179 9,平均为1.435 2。同时,基于SSR分析结果得到编码各异的品种分子身份证,该分子编码可以将亲缘关系不同的欧李品种进行有效区分;聚类分析显示19个欧李品种及其近缘种根据亲缘关系远近聚为不同类群。结果说明,SSR标记在蔷薇属种间具有通用性,且是欧李种质鉴定及其分子身份证构建的有效手段。
【Abstract】 The molecular ID of 19 varieties of Cerasus humilis and its closely related plant species were constructed by fluorescent SSR markers. A total of 13 pairs of SSR primers with high polymorphism and good stability were selected from its closely related plant species, and used for analysis of genetic diversity and genetic relationship of C. humilis, Prunussimonii and P. salicina Lindl. A total of 88 alleles were detected, the number of alleles per pair of SSR primer was 2-12 with an average of 6.7. Ninty-eight bands were obtained ranging in 3-12 with an average of 7.5. The polymorphic information content ranged in 0.296 1-0.853 3 with an average of 0.629 7, and the Shannon’s information index ranged iin 0.547 4-2.179 9 with an average of 1.435 2. From the SSR analysis results, the molecular IDs of different varieties of codes are obtained. The molecular code can effectively distinguish C. humilis varieties with different genetic relationships. By the cluster analysis, the 19 C. humilis varieties and their related species are clustered into different groups according to their close relationship. Using SSR markers is versatile among Rosa species, and is an effective method for germplasm identification and molecular ID cards construction of C. humilis.
