基于环介导等温扩增技术的巴戟天检测方法的建立

【摘要】 目的建立一种可快速检测中药巴戟天的环介导等温扩增(LAMP)检测方法。方法针对巴戟天二级内部转录空间(ITS2)序列设计LAMP特异引物组,提取DNA后建立基于LAMP技术鉴别巴戟天的目视和实时定量检测方法,同时评估该方法的特异性、灵敏度及其在实践中的应用情况。结果该检测方法具有良好的特异性,最适反应温度为63℃,灵敏度约为0.10μg/L,比常规聚合酶链式反应(PCR)方法约高10倍。经验证可成功从7种近缘物种中检测出巴戟天。结论巴戟天LAMP检测是一种简便、客观、灵敏、快速的方法,值得进一步推广应用。

【Abstract】 Objective To establish a loop-mediated isothermal amplification(LAMP) method for rapid detection of the traditional Chinese medicine Morinda officinalis How. Methods LAMP specific primers were designed for the second internal transcriptional space(ITS2) sequence of Morinda officinalis How, and after DNA extraction, a visual and real-time quantitative detection method was established for the identification of Morinda officinalis How based on LAMP technique. The specificity, sensitivity, and application of this method in practice were evaluated. Results This detection method had good specificity, an optimum reaction temperature of 63 ℃, and a sensitivity of 0.10 μg/L, which was about 10 times higher than conventional polymerase chain reaction. Validation showed that Morinda officinalis How was successfully detected from 7 related species. ConclusionLAMP is a simple, objective, sensitive, and rapid detection method for Morinda officinalis How and thus holds promise for further application.
