HPLC法同时测定核桃楸枝皮中四种植物激素
Simultaneous Determination of Four Phytohormones in the Bark of Juglansmandshuricaby HPLC
【摘要】 为建立同时测定核桃楸枝皮中4种内源激素3-吲哚乙酸(IAA)、3-吲哚丁酸(IBA)、萘乙酸(NAA)和脱落酸(ABA)含量的高效液相色谱法方法,本文对供试样品制备方法和高效液相色谱条件进行了优化,并对其进行了验证。结果表明:最优的测定样品制备方法是超声提取法;色谱条件为HIQ Sil C18W色谱柱(250mm×4.6mm,5μm);以0.06%甲酸水为流动相A,甲醇为流动相B,进行梯度洗脱;检测波长为280nm;流速为1.0mL·min-1;柱温为25℃;进样量20μL。方法学验证结果表明,利用高效液相色谱法同时测定核桃楸枝皮中IAA、IBA、NAA和ABA四种内源激素的方法是可行的。测得核桃楸枝皮中IAA、IBA、NAA和ABA的含量分别为39.87,32.50,34.60和65.23ng·g-1。该方法操作快捷、重现性好、灵敏度高,适用于核桃楸枝皮中激素含量分析。
【Abstract】 In order to establish an HPLC method for the simultaneous determination of four endogenous hormones in the bark of Juglans mandshurica,including 3-indoleacetic acid(IAA),3-indolebutyric acid(IBA),naphthylacetic acid(NAA)and abscisic acid(ABA),the sample preparation method and HPLC conditions were optimized.The results showed that the optimal sample preparation method was ultrasonic extraction;chromatographic conditions:HiQ SIL C18 W column(250 mm ×4.6 mm,5μm);mobile phase:0.06%formic acid water as mobile phase A,methanol as mobile phase B,gradient elution;detection wavelength:280 nm;flow rate:1.0 mL·min-1;column temperature:25 ℃;injection volume:20μL.The results showed that the method was feasible for the simultaneous determination of IAA,IBA,NAA and ABA in the bark of Juglans mandshurica.The contents of IAA,IBA,NAA and ABA were 39.87,32.50,34.60 and 65.23 ng·g-1,respectively.The method is rapid,reproducible and sensitive,which is suitable for the analysis of hormone content in bark of Juglans mandshurica.
