血清外泌体对乳腺癌细胞生物学行为的影响
Effects of serum exosomes on biological behavior of breast cancer cells
【摘要】 目的:探索血清外泌体对乳腺癌细胞株增殖、迁移、凋亡和化疗药敏感性的影响。方法:利用超速离心法从治疗效果良好的乳腺癌患者血清中分离外泌体,并通过透射电镜观察、粒径分析、流式细胞术、蛋白印迹法进行鉴定。将外泌体与乳腺癌细胞株MCF-7共同培养,以未处理的MCF-7细胞为对照,分别通过CCK-8法、划痕实验、流式细胞术分析外泌体对MCF-7细胞增殖、迁移、凋亡、阿霉素敏感性的影响。结果:通过超速离心可分离得到合格且足量的外泌体。与对照组比较,实验组细胞的增殖能力明显被抑制(P<0.05);实验组细胞的划痕愈合率显著降低(P<0.01);实验组细胞有较高的凋亡率(P<0.05)。与对照组比较,外泌体与阿霉素联合作用对细胞增殖的抑制能力更强(P<0.05),但外泌体与Taxol联合对细胞的抑制作用无明显变化。结论:疗效良好的乳腺癌患者血清外泌体可有效抑制乳腺癌细胞的增殖和迁移,并诱导凋亡以及增强阿霉素的敏感性。
【Abstract】 Objective: To explore the effects of serum exosomes on proliferation, migration, apoptosis and chemosensitivity of breast cancer cell lines. Methods: Exosomes were isolated from the serum of breast cancer patients with good therapeutic effect by ultracentrifugation, and identified by transmission electron microscopy, particle size analysis, flow cytometry and Western blot. The effects of exosomes on the proliferation, migration,apoptosis and adriamycin sensitivity of MCF-7 cells were analyzed by CCK-8 method, scratch test and flow cytometry, respectively. Results: Qualified and sufficient exosomes were obtained using ultracentrifugation. The proliferation of the cells in the study group was significantly inhibited(P<0.05). The scratch healing rate of cells in the study group was significantly reduced, and while the apoptosis rate was elevated compared with the control group(P<0.05). The combination of exosomes and adriamycin had stronger inhibition on cell proliferation(P<0.05). However, the combination of exosomes and taxol had no significant effect on cell proliferation. Conclusion: The serum derived exosomes from breast cancer patients with good therapeutic effect can effectively inhibitthe proliferation and migration of breast cancer cells,induce apoptosis, and enhance the sensitivity of adriamycin.
