宁夏西瓜、甜瓜白粉病病原菌鉴定及抗性种质资源筛选

【摘要】 【目的】针对宁夏特色优势作物西瓜、甜瓜白粉病发生日趋严重的问题.【方法】采用镜检法、ITS序列分子鉴定和国际通用的甜瓜白粉病生理小种鉴别寄主与标准鉴定体系,对宁夏地区不同地点、不同栽培方式的西瓜、甜瓜上采集的11份病菌样本进行白粉病病原菌及生理小种鉴定,并采用离体叶片鉴定方法对23份甜瓜种质资源进行抗病性鉴定.【结果】11份病菌样本的分生孢子形态符合单囊壳白粉菌的形态特征,有纤维状体,病菌样本的rDNA-ITS序列与单囊壳白粉菌Podosphaera xanthii的ITS序列聚为一类,病菌样本对鉴别寄主的抗感反应与瓜单囊壳白粉菌生理小种1表现一致.【结论】引起宁夏地区西瓜、甜瓜白粉病的病原菌为单囊壳白粉菌(Podosphaera xanthii),且其生理小种为小种1,未发现其他生理小种.筛选出对白粉病免疫的材料有1489(发病率和病情指数均为0),高抗材料有1464、Sweet和抗病F-3800共3份(发病率低且病情指数均在10以下).

【Abstract】 【Objective】 In order to identify the pathogen of powdery mildew of watermelon and muskmelon in Ningxia, and screen the resistance germplasm resources.【Method】 The pathogens and physiological races of 11 powdery mildew samples collected from the watermelon and muskmelon planted in different cultivation methods in different sites of Ningxia were identified, by using microscopic examination, ITS sequence molecular identification, international common physiological races identification hosts of melon powdery mildew and its standard identification system, and the disease resistance of 23 melon germplasm resources was identified by in vitro leaf identification method. 【Result】 The conidia morphology of 11 pathogen samples was consistent with that of powdery mildew with fibrous bodies. The rDNA-ITS sequence of the pathogen sample was clustered with the ITS sequence of powdery mildew Podosphaera xanthii. The resistant and susceptible response of the pathogen samples to the identified host was consistent with that of race 1 resistance responses of the pathogen samples to identification hosts were consistent with that of physiological race 1 of Podosphaera xanthii. 【Conclusion】 The pathogen causing powdery mildew of watermelon and melon in Ningxia was identified as Podosphaera Xanthii,and its physiological race was race 1,no other physiological race was found. The material 1489 was the one which immune to powdery mildew, with incidence and disease index of 0,1464,Sweet and F-3800 were the three high resistant materials, with low incidences and below 10 disease indexes.
