杂交籼稻稻瘟病抗性分子标记辅助育种的探索与实践
Exploration and practice on improvement of indica hybrid rice blast resistance by using molecular marker assisted breeding
【摘要】 利用具有广谱和持久抗性的抗稻瘟病基因培育抗病品种是控制稻瘟病危害最有效途径之一。通过分子标记辅助选择将广谱抗稻瘟病基因Pigm导入温敏核不育系隆科638S,育成新的抗稻瘟病温敏核不育系扬籼6S;利用分子标记将华占的稻瘟病抗性基因Pi2、Pi54、Pid3、Pita和Pib等导入辐恢838中,育成新的抗稻瘟病恢复系扬恢1512,与华占相比,抗性提高,结实率和千粒重显著增加。以扬籼6S作母本,以扬恢1512作父本,配制杂交组合,育成两系杂交籼稻新组合扬两优612,其稻瘟病抗性综合指数为2.8,表现中抗。扬籼6S和扬恢1512的成功选育,为培育抗稻瘟病杂交稻新品种提供了抗病新种质;而扬两优612的成功选育,则为杂交水稻稻瘟病抗性育种实践提供了实例。
【Abstract】 Using broad-spectrum and durable resistance genes in rice breeding is one of the most effective ways to control rice blast disease in rice production. In this study, the broad-spectrum blast resistance gene Pigm was introduced into the thermo-sensitive male sterile(TMS) line Longke 638 S by using molecular marker-assisted selection, and a new blast resistance TMS line Yangxian 6 S was developed. The Pi2, Pi54, Pid3, Pita and Pib from Huazhan were introduced into Fuhui 838 by using molecular marker-assisted selection and a new rice blast resistance restorer line Yanghui 1512 was bred. Compared to Huzhan, Yanghui 1512 displayed higher resistant level to blast disease, especially the seed setting rate and thousand-grain weight were increased significantly. Using Yangxian 6 S as the female parent and Yanghui 1512 as the male parent, we produced a new two-line hybrid indica rice combination Yangliangyou 612, which had a comprehensive blast resistance index of 2.8 and reached medium resistance level. The successful breeding of Yangxian 6 S and Yanghui 1512 provided new resistance germplasm for breeding new rice blast resistant hybrid varieties, and the successful breeding of Yangliangyou 612 provided breeding practice examples for developing blast resistant hybrid rice.
