不同黑臭程度水体底泥细菌群落多样性及组成分析
Analysis of diversity and composition of bacterial community in sediments under different black-odor levels
【摘要】 采用高通量测序技术对不同黑臭程度水体底泥细菌群落结构进行测定,分析底泥理化因子与细菌群落组成的关系。结果表明:不同黑臭程度水体底泥细菌优势菌门均由Proteobacteria、Chloroflexi和Bacteroidetes组成,其中重度黑臭水体底泥Proteobacteria相对丰度高于轻度黑臭水体底泥,而Bacteroidetes呈相反趋势;黑臭水体底泥中存在丰富的潜在除臭菌,在重度黑臭水体底泥中相关功能菌总相对丰度高于轻度黑臭水体;重度黑臭水体底泥中细菌群落多样性明显较低且相似性较差。冗余分析(RDA)及相关性分析表明,不同黑臭程度水体底泥细菌群落α-多样性与底泥铵态氮、总氮、有机质含量呈显著负相关,而与总磷、溶解氧含量表现出正相关关系,溶解氧是影响Proteobacteria和Bacteroidetes相对丰度的主要因素。
【Abstract】 Bacterial community in sediments under different black-odor levels was evaluated using Illumina MiSeq sequencing of the bacterial 16 S rRNA genes. The results showed that the predominant phyla in sediments were Proteobacteria, Chloroflexi and Bacteroidetes, and the relative abundance of proteobacteria in sediment under severe black-odor water was higher than that of mild black-odor water, while Bacteroidetes showed the opposite trend. In addition, the high proportion of deodorant bacterial was found in sediments under different black-odor levels, including Halomonas, Pseudomonas, Thiobacillus, Sulfuricurvum, and the relative abundances of these deodorant bacterial showed different trends in sediments under different black-odor levels. From the perspective of community diversity, the Shannon index in sediment under severe black-odor water was significantly lower than that of mild black-odor water, and had a low similarity of bacterial communities. Redundancy analysis(RDA) and correlation analysis showed that the composition and diversity of bacterial community in sediments under different black-odors water were affected by nutrients and dissolved oxygen.
