超声提取-分光光度法测定巴戟天中总蒽醌含量

【摘要】 建立了超声提取-分光光度法测定巴戟天中总蒽醌含量的方法。样品经粉碎处理后,用质量分数为80%乙醇作溶剂,在超声下提取,提取液加盐酸水解后以氯仿萃取总蒽醌,最后将氯仿挥发干后,用醋酸镁-甲醇法测定总蒽醌含量。探讨了超声提取及后续处理条件的优化过程,并与传统的加热回流提取法进行了比较。结果表明,新方法操作简便、结果准确,对实际样品测定的相对标准偏差为0.75%~5.65%(n=3),回收率为94.00%~104.98%,相对于加热回流提取法,提取率提高了2倍以上。该方法可用于中药企业产品质量控制检测,具有较高的应用价值。

【Abstract】 A method was established for determining the content of total anthraquinone in Morindae officinalis Radix by ultrasonic extraction and spectrophotometry. The sample was crushed and extracted under ultrasonication with ethanol aqueous solution(80%) as solvent. Total anthraquinone was extracted by chloroform from the extraction which was hydrolyzed with hydrochloric acid in advance. The content of total anthraquinone was determined by magnesium acetate-methanol spectrophotometry after the removal of chloroform. The optimal conditions of ultrasonic extraction and subsequent treatment were mainly discussed, and the data of ultrasonic extraction were compared to those of traditional heating reflux extraction. The results showed that this new approach was simple and accurate. Its relative standard deviations for determining actual samples were between 0.75% and 5.65%(n=3) and recovery rates were between 94.00% and 104.98%. In comparison with heating reflux extraction, the extraction rate of new method increased by more than two times. This new method can be used for product quality inspection in traditional Chinese medicine enterprise with high application value.
