miR-141-3p在前列腺癌患者血清中的表达及对骨转移的调控机制

【摘要】 目的研究分析miR-141-3p在前列腺癌患者血清中的表达水平以及对骨转移的调控机制。方法选取2017年1月至2019年1月于我院肿瘤科就诊的85例前列腺癌患者作为研究对象,根据患者的疾病分类进行分组,其中45例局限性前列腺癌患者作为观察A组,40例激素敏感性骨转移前列腺癌作为观察B组,并选取同期于我院诊断的43例良性前列腺增生患者作为对照组,比较观察A组、观察B组以及对照组患者miR-141-3p血清中的表达水平;并将观察A组与观察B组85例前列腺癌患者按照骨转移灶个数进行再次分组,分为无骨转移灶、0~2个骨转移灶、3~5个骨转移灶以及5个以上骨转移灶组,分析4组患者血清中miR-141-3p-3p与前列腺骨转移的相关性;并根据人前列腺癌细胞株PC-3与正常小鼠前列腺组织、小鼠前列腺癌骨转移组织进行分析miR-141-3p的潜在靶基因对前列腺癌骨转移的调控。结果与对照组比较,观察A、B组患者前列腺癌组织miR-141-3p表达水平差异有统计学意义(P<0.05),且观察A组明显高于观察B组,差异有统计学意义(P<0.05);观察A、B组患者血清中miR-141-3p表达水平明显高于对照组,差异有统计学意义(P<0.05),且观察A组明显高于观察B组,差异有统计学意义(P<0.05)。血清中miR-141-3p表达水平与前列腺骨转移个数呈正相关关系(r=0.517,P<0.05);人前列腺癌细胞株DU145、PC-3中,miR-141-3p mimics组miR-141-3p表达水平高于miRNA-NC组,差异有统计学意义(P<0.05);miR-141-3p可诱导人前列腺癌细胞株DU145、PC-3侵袭能力增加;人前列腺癌细胞株DU145、PC-3中,miR-141-3p mimics组IL11表达水平低于miRNA-NC组,差异有统计学意义(P<0.05);正常小鼠前列腺组织与前列腺癌细胞株中IL11与miR-141-3p表达水平呈负相关关系(r=-0.644,P<0.05);小鼠前列腺癌骨转移组织中IL11与miR-141-3p表达水平呈负相关关系(r=-0.580,P<0.05)。结论 miR-141-3p的血清表达可作为前列腺早期诊断治疗的生物学标志物,且miR-141-3p可作为患者骨转移的分子标记物,其miR-141-3p可通过IL11完成对前列腺癌骨转移的调控。

【Abstract】 ObjectiveTo analyze the expression level of mi R-141-3 p in serum of patients with prostate cancer andits regulation mechanism on bone metastasis.MethodsEighty-five patients with prostate cancer admitted to our oncologydepartment from January 2017 to January 2019 were enrolled in the study. According to the patient′s disease classification,45 patients with localized prostate cancer were used as observation group A,40 patients with hormone-sensitive bone metas-tases prostate cancer as observation group B,and 43 patients with benign prostatic hyperplasia diagnosed in our hospital ascontrol group. The expression levels of mi R-141-3 p serum in observation group A,group B and control group were compared.Eighty-five patients with prostate cancer in observation group A and observation group B were regrouped according to thenumber of bonemetastase lesions,including4 groups of nobone metastases,0~2 bone metastases,3~5 bone metastases,andover 5 bone metastases. The correlation between mi R-141-3 p-3 p and prostate bone metastasis in the serum of the four groupswas analyzed.According to the human prostate cancer cell line PC-3,normal mouse prostate tissue,and mouse prostatecancer bone metastasis tissue,the potential target genes of mi R-141-3 p were analyzed for the regulation of prostate cancerbone metastasis.ResultsThe expression levels of mi R-141-3 p in prostate cancer tissues of group A and group B were signifi-cantly higher than those of control group,and group A was higher than group B. The difference was statistically significant(P<0.05);The expression levels of mi R-141-3 p in serum of patients in group A and group B were significantly higher thanthose in control group,and group A was higher than group B. The difference was statistically significant(P<0.05);Serummi R-141-3 p expression level was positively correlated with prostate bone metastasis(r=0.517,P<0.05). In human prostatecancer cell lines DU145 and PC-3,mi R-141-3 p mimics group mi R-141-3 p expression level was higher than mi RNA-NCgroup,the difference was statistically significant(P<0.05). mi R-141-3 p can induce increased invasion ability of human pros-tate cancer cell lines DU145 and PC-3. In human prostate cancer cell lines DU145 and PC-3,the IL11 expression level of mi R-141-3 p mimics group was lower than that of mi RNA-NC group,the difference was statistically significant(P<0.05). Theexpression levels of IL11 and mi R-141-3 p in normal mouse prostate tissues and prostate cancer cell lines were negativelycorrelated(r=-0.644,P<0.05). There was a negative correlation between IL11 and mi R-141-3 p expression level(r=-0.580,P<0.05).ConclusionThe serum expression of mi R-141-3 p can be used as a biomarker for early diagnosis and treatment ofprostate,and mi R-141-3 p can be used as a molecular marker for bone metastasis in patients,which can regulate the bonemetastasis of prostate cancer through IL11.
