大豆种子分泌物中蛋白质鉴定及其对大豆疫霉的趋化作用

【摘要】 研究以大豆抗、感品种接种大豆疫霉前后种子分泌物中蛋白质为研究对象,测定其对大豆疫霉游动孢子趋化性、成囊和萌发的影响,在此基础上采用LC/MS-MS鉴定蛋白质种类。体外活性试验表明,大豆抗病品种种子分泌物中总蛋白质排斥大豆疫霉游动孢子、抑制游动孢子成囊和萌发,且接种后抑制作用更强。大豆感病品种接种前种子分泌物中总蛋白质吸引大豆疫霉游动孢子,接种后转为排斥。大豆抗、感品种接种前种子分泌物中共鉴定得到多聚半乳糖醛酸酶抑制剂、类kiwellin蛋白、Bowman-Birk型蛋白酶抑制剂、类过氧化物酶等17种已报道具有植物防御活性差异蛋白,抗病品种防御蛋白种类多于感病品种;大豆抗病品种接种后诱导表达超氧化物歧化酶、PR-4、β-1, 3-葡聚糖内切酶等9种已报道与植物防御相关差异蛋白,大豆感病品种接种后诱导表达类PR-5、类α-淀粉酶/枯草杆菌蛋白酶抑制剂等8种具有防御活性差异蛋白。除已知具有防御活性的蛋白质外,其余诱导表达的差异蛋白也可能参与防御过程,在种子萌发过程中防御土壤中病原物包括大豆疫霉的侵染。

【Abstract】 In this study, the effects of proteins in the seed exudates of soybean resistance and susceptible cultivars before and after inoculation of Phytophthora sojae on zoospore chemotaxis, formation of cyst and cyst germination of P. sojae were determined, the proteins were identified by LC/MS-MS. In vitro activity tests showed that the proteins in the seed exudates of resistance soybean did not attract zoospores,inhibited the formation of cyst and cyst germination of P. sojae, and the inhibition became stronger after inoculation. The proteins in the seed exudates of soybean susceptible cultivar attracted zoospores before inoculation but repelled zoospores after inoculation. Both of resistance and susceptible soybean seed exudates before inoculation were identified with 17 differential proteins which were reported to be related with plant defense, including polygalacturonase inhibitor protein, kiwellin-like protein, Bowman-Birk type proteinase inhibitor, peroxidase-like protein and so on. The proteins in the seed exudates of the resistance cultivar were more than those in the susceptible cultivar. After inoculation, the resistance soybean seeds were induced to express nine differential proteins reported to be related to plant defense, including PR-4, endo-1, 3-beta-glucanase and so on. Eight differential proteins in the seed exudates of the susceptible soybean with defensive activity were induced after inoculation, such as PR-5 like protein, alpha-amylase/subtilisin inhibitor-like protein. In addition to the certain proteins with defensive activity, other differentially expressed proteins might also involve in the defense process to defense soil pathogens, including P. sojae, during the seed germination stage.
