紫薇属种间杂种SSR分子鉴定与遗传分析

【摘要】 以紫薇品种及优良无性系为母本,以大花紫薇和福建紫薇为父本进行杂交,随机选择ZD(紫薇×大花紫薇)和ZF(紫薇×福建紫薇)2个杂种群体51个优良单株为试材,从51对SSR引物中筛选出15对在亲本间具多态性的引物对子代进行真实性鉴定,以遗传相似系数和遗传距离对亲子代个体进行UPGMA聚类分析。结果表明:ZD和ZF 2个群体各19个单株被至少2对引物鉴定为真杂种,群体平均等位基因数(Na)分别为4.6和5.0,平均期望杂合度(He)为0.573 2和0.657 3,平均Shannon′s信息指数(I)为1.044 5和1.215 2,平均多态信息含量(PIC)为0.504 7和0.586 0,说明2个子代群体遗传多样性较高。亲子代间的遗传相似系数在0.62~1.00,存在遗传差异。聚类树结果显示ZD群体与母本紫薇聚在一起,亲缘关系较近,ZF群体则与父本福建紫薇亲缘关系较近。

【Abstract】 Lagerstroemia indica used as the female parents were crossed with the L.speciosa and L.limii as the male parents,51 excellent individuals were randomly selected from the ZD and ZF hybrid progeny populations were used as test materials.In this study,15 pairs of primers were found polymorphic within parents among 51 pairs of SSR primers that were used to confirm the hybrids.Based on genetic distance and genetic similarity,UPGMA cluster analysis was performed on the parents and offspring.The results showed that 19 individuals in each of the ZD and ZF populations were proved to be the true hybrid by at least two markers.The mean value of number of alleles(Na)were 4.6 and 5.0.The mean value of expected heterozygosity(He) were 0.573 2 and 0.657 3.The average Shannon′s information index(I) were 1.044 5 and 1.215 2.And the average value of polymorphism information content(PIC) were 0.504 7 and 0.586 0.The above results indicated that both ZD and ZF had higher genetic diversity.The genetic similarity ranged from 0.62 to 1.00 between these hybrids and their parents which showed genetic differentiation.The UPGMA dendrogram tree showed that ZD offspring were closely related to L.indica,and ZF offspring were closely related to L.limii.
