【摘要】 目的:研究重楼皂苷VII对大鼠骨关节炎和细胞外基质影响。方法:采用前交叉韧带横断(ALCT)手术处理大鼠,将成模大鼠分为:骨关节炎模型组、重楼皂苷VII 25 mg/kg、50 mg/kg、100 mg/kg组和双醋瑞因胶囊50 mg/kg组,另设假手术组。开放性运动测试检测大鼠10 min运动距离;苏木素-伊红染色和番红O-固绿染色检测大鼠骨关节病理损伤程度;ELISA试剂盒检测膝关节液中肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、白介素1β(IL-1β)水平和血清中IL-1β、基质金属蛋白酶9(MMP9)、解聚素金属蛋白酶5(ADAMTS-5)含量。结果:与假手术组相比较,骨关节炎模型组10 min运动距离显著降低,病理损伤程度明显改善且Mankin’s评分及OARSI评分显著升高,膝关节液中TNF-α、IL-6、IL-1β水平显著升高,血清中IL-1β、MMP9、ADAMTS-5含量显著升高;与骨关节炎模型组相比较,重楼皂苷VII 50、100 mg/kg和双醋瑞因胶囊组10 min运动距离显著升高,病理损伤程度明显改善且Mankin’s评分及OARSI评分显著降低,膝关节液中TNF-α、IL-6、IL-1β水平显著降低,血清中IL-1β、MMP9、ADAMTS-5含量显著降低。结论:重楼皂苷VII具有改善骨关节炎病理损伤程度,调节细胞外基质的作用。更多还原

【Abstract】 Objective:To investigate the effects of Polyphyllin VII on pathological injury and extracellular matrix in rats with osteoarthritis. Methods:Osteoarthritis rat model was established by anterior cruciate ligament transversal(ALCT) surgery and were divided into the sham surgery group,the model group(Osteoarthritis, OA), Polyphyllin VII groups(25 mg/kg、50 mg/kg、100 mg/kg) and Diacerein Capsules group(50 mg/kg). The movement distance of the rats was measured in open motion test for 10 minutes. Hematoxylin-eosin staining and saffir O solid green staining were used to detect the degree of pathological injury in bone and joint. The contents of TNF-a、IL-6、IL-1β in Knee joint fluid and serum IL-1β, MMP9、ADAMTS-5 were detected by ELISA. Results:Compared with the model group, the movement distance of osteoarthritis rats was significantly decreased in the model group, the pathological injury of bone and joint tissue was improved, scores of Mankin’s and OARSI were increased, levels of TNF-α、IL-6 and IL-1βin knee joint fluid were increased, levels of IL-1β、MMP9、ADAMTS-5 in serum were significantly increased. Compared with the model group, the movement distances of osteoarthritis rats were increased in Polyphyllin VII groups(50、100 mg/kg) and 50 mg/kg Diacerein group, Also, the pathological injury was improved, scores of Mankin’s and OARSI were significantly reduced, levels of TNF-α、IL-6 and IL-1βin knee joint fluid were decreased, levels of IL-1β、MMP9、ADAMTS-5 in serum were significantly decreased.Conclusion: Polyphyllin VII can improve the pathological damage of osteoarthritis and regulate extracellular matrix.更多还原
