基于EST-SSR标记的茅苍术种质资源遗传多样性分析

【摘要】 采用EST-SSR标记,对5种不同叶型47份茅苍术进行遗传多样性分析。从30对引物中筛选出5对EST-SSR引物用于PCR扩增,共扩增出36条多态性条带,遗传多态性PIC值变化范围为0.6760~0.7877,平均为0.7539;供试样品遗传距离为0~0.9123,表明茅苍术具有丰富的遗传多样性。采用UPGMA法进行聚类分析,当遗传相似系数为0.70时,可将供试样品分为5大类。聚类结果显示叶片形态特征和分子表达并不一致,即茅苍术的叶形性状与遗传背景不具有明显的相关性。

【Abstract】 Genetic diversity analysis on fourty-seven samples of 5 leaf form of Atractylodeslancea(Thunb.) D C. using EST-SSR markers. Five pairs of primer which were screened from thirty pairs had been applied in PCR amplification. A total of 36 bands had been amplified, 36 of which are polymorphic bands, accounted for 100%. The PIC varied with between 0.6760~0.7877(0.7539 on average). Genetic distance of samples was 0~0.9123, show that A. lanceais highly diverse. The samples were indentified into five types by UPGMA when the genetic distance was 0.70. The results of clustering show that there was no relationship between leaf characters and molecular expression, that there was no obvious relativity between leaf characters and genetic background of A. lancea.
