外周血DNT细胞在自身免疫性疾病中的表达及意义

【摘要】 目的研究外周血双阴(double positive, DN)T细胞在自身免疫性疾病患者的分布特点,以及自身免疫性疾病患者经过免疫治疗后DNT细胞的变化情况。方法选择首次确诊未经治疗的38例自身免疫性疾病患者作为治疗前组,经过激素和或激素联合免疫抑制剂治疗后满4周的13例自身免疫性疾病患者作为治疗后组,同期30例健康体检者作为对照组。利用流式细胞术检测三组研究对象的外周血DNT细胞含量,比较三组研究对象外周血DNT细胞含量。结果治疗前组患者外周血DNT细胞含量为(4.93±2.78)%,高于对照组的(3.90±1.90)%,但差异无统计学意义(P>0.05);治疗后组患者外周血DNT细胞含量为(2.20±0.80)%,低于治疗前组的(4.93±2.78)%,差异具有统计学意义(P<0.05)。结论自身免疫性疾病患者外周血DNT细胞含量高于健康人群,说明DNT细胞有免疫促进作用,自身免疫性疾病患者经过免疫治疗后DNT细胞较治疗前明显下降,说明DNT细胞参与下调免疫反应的作用。

【Abstract】 Objective To study the distribution characteristics of double negative(DN) T cells in peripheral blood in patients with autoimmune diseases and the changes of DNT cells in patients with autoimmune diseases after immunotherapy. Methods There were 38 patients with autoimmune diseases diagnosed for the first time without treatment selected as the pre-treatment group, 13 patients with autoimmune diseases after 4 weeks of treatment with hormone or hormone combined immunosuppressant selected as the post-treatment group, and 30 healthy persons in the same period as the control group. Flow cytometry was used to detect the content of DNT cells in the peripheral blood of the three groups, and the content of DNT cells in the peripheral blood of the three groups was compared. Results The content of DNT cells in the peripheral blood in pre-treatment group was(4.93±2.78)%, which was higher than(3.90±1.90)% in the control group, but the difference was not statistically significant(P>0.05). The content of DNT cells in the peripheral blood in post-treatment group was(2.20±0.80)%, which was lower than(4.93±2.78)% in pre-treatment group, and the difference was statistically significant(P<0.05). Conclusion The content of DNT cells in the peripheral blood of the patients with autoimmune diseases is higher than that of the healthy people, which indicates that DNT cells have immune promoting effect. After immunotherapy, DNT cells in the patients with autoimmune diseases are significantly lower than that before treatment, which indicates that DNT cells participate in the down-regulation of immune response.
