黄芪匀浆膳通过NLRP3炎症小体通路影响创伤应激大鼠肠黏膜免疫功能
Effects of Astragalus homogenized diet on intestinal mucosal immune function and NLRP3 inflammatory corpuscle pathway of traumatic stress rats
【摘要】 目的:探讨黄芪匀浆膳对创伤应激大鼠肠黏膜免疫功能及NLRP3炎症小体通路的影响。方法:按照随机数字表法将大鼠分为5组:空白对照组、模型组、黄芪匀浆膳组、NLRP3激活剂组、NLRP3激活剂+黄芪匀浆膳组,每组12只。采用双侧股骨离断及胃内硅胶管置入建立创伤大鼠模型。流式细胞仪检测小肠黏膜固有层淋巴细胞亚群CD3+、CD4+、CD8+和CD19+,酶联免疫法(ELISA)检测肠黏膜组织SIg A、IL-2、IL-4、IL-6、TGF-β、IL-1β、IL-18浓度,实时荧光定量PCR(RT-PCR)和Western blot分别检测NLRP3、ASC、cleaved-caspase-1 mRNA和蛋白表达。结果:与空白对照组比,模型组CD3~+、CD4~+、CD4~+/CD8~+、CD19~+、SIg A、IL-2、IL-4、IFN-γ和TGF-β显著降低(P<0. 05),IL-6、IL-1β、IL-18、NLRP3、ASC、cleaved-caspase-1 mRNA和蛋白表达均显著升高(P<0. 05)。与模型组相比,黄芪匀浆膳组CD3+、CD4+、CD4+/CD8+、CD19+、SIg A、IL-2、IL-4、IFN-γ和TGF-β显著升高(P<0. 05),IL-6、IL-1β、IL-18、NLRP3、ASC、cleaved-caspase-1 mRNA和蛋白表达均显著降低(P<0. 05)。加入NLRP3激活剂后,黄芪匀浆膳促进CD3+、CD4+、CD19+、SIg A、IL-2、IL-4、IFN-γ和TGF-β表达,降低IL-6、IL-1β、IL-18、NLRP3、ASC、cleaved-caspase-1 mRNA和蛋白表达。结论:黄芪匀浆膳可能通过抑制NLRP3炎症小体通路的激活提高创伤应激大鼠肠黏膜的免疫功能。
【Abstract】 Objective: To investigate the effects of astragalus homogenized diet on intestinal mucosal immune function and NLRP3 inflammatory corpuscle pathway of traumatic stress rats. Methods: The rats were randomly divided into 5 groups according to the random number table: blank group,model group,Astragalus homogenized diet group,NLRP3 activator group,NLRP3 activator~+Astragalus homogenized diet group,with 12 rats in each group. The traumatic rat model was established by bilateral femur dissection and intragastric silicone tube insertion. Lymphocyte subsets CD3~+,CD4~+,CD8~+and CD19~+in intestinal mucosa lamina propria were detected by flow cytometry,the concentrations of SIg A,IL-2,IL-4,IL-6,TGF-β,IL-1β and IL-18 in intestinal mucosa tissues were detected by enzyme-linked immunosorbent assay( ELISA),real-time fluorescence quantitative PCR( RT-PCR) and Western blot were used to detect the expressions of NLRP3,ASC,cleaved-caspase-1 mRNAs and proteins respectively. Results: Compared with the blank control group,CD3~+,CD4~+,CD4~+/CD8~+,CD19~+,SIg A,IL-2,IL-4,IFN-γ and TGF-β in model group were significantly decreased( P<0. 05),IL-6,IL-1β,IL-18,NLRP3,ASC,cleaved-caspase-1 mRNA and protein expression were significantly increased( P < 0. 05). Compared with the model group,CD3~+,CD4~+,CD4~+/CD8~+,CD19~+,SIg A,IL-2,IL-4,IFN-γ and TGF-β in astragalus homogenized diet groups were significantly increased( P<0. 05); IL-6,IL-1β,IL-18,NLRP3,ASC,cleaved-caspase-1 mRNA and protein expression were decreased significantly( P<0. 05). After adding NLRP3 activator,Astragalus homogenate diet promoted the expression of CD3~+,CD4~+,CD19~+,SIg A,IL-2,IL-4,IFN-γ,TGF-β and decreased the expression of IL-6,IL-1β,IL-18,NLRP3,ASC,cleaved-caspase-1 mRNA and protein. Conclusion: Astragalus homogenized diet may enhance the immune function of intestinal mucosa in traumatic stress rats by inhibiting the activation of NLRP3 inflammatory corpuscle pathway.
