利用SSR构建甜菜品种的分子身份证
Establishment of Molecular Identity for Sugar Beet Cultivars based on SSR Markers
【摘要】 为了实现快速鉴定甜菜品种真实性以及纯度的目的,本研究利用SSR分子标记技术对国内已登记的部分甜菜品种进行了指纹图谱构建。利用17对带型清晰、重复性好、多态性高、易于识别的引物作为核心引物对39份甜菜品种的基因组DNA进行扩增。结果表明,只需利用以下5对核心引物:LNX38、LNX95、SB06、SSD6和SB13,便可完全鉴别出39个甜菜品种。利用SSR引物构建甜菜品种的分子身份证,真正达到了快速、高效地鉴定品种纯度以及真实性的目的,也更有效地维护了甜菜品种市场的秩序,并保护了育种家的利益。
【Abstract】 In order to identify the authenticity and purity of sugar beet varieties rapidly, the SSR molecular marker technique was used to construct the fingerprint of the registered sugar beet varieties in China. In the experiment, 17 pairs of primers with clear pattern, good repeatability, high polymorphism and easy recognition were found through the detection of 200 pairs of SSR amplification products. The 17 pairs of primers were used as core primers to amplify the genomic DNA of 39 sugar beet varieties, and the SSR fingerprints of 39 sugar beet varieties were successfully constructed. After that, the phylogenetic tree map of the all varieties was constructed through the data of fingerprinting. It was found that 39 sugar beet varieties could be completely identified by using the following 5 pairs of core primers: LNX38, LNX95, SB06, SSD6 and SB13. Using SSR primers to construct fingerprinting for sugar beet varieties makes us truly achieve the goal of rapid and efficient identification of variety purity and authenticity,which also more effectively maintains the market order of sugar beet variety, and protects the interests of breeders.
