侧脑室微量注射神经肽W30对清醒大鼠下丘脑室旁核区谷氨酸含量的影响

【摘要】 [目的]探讨侧脑室微量注射给予神经肽W30(NPW30)对清醒大鼠下丘脑室旁核(PVN)区谷氨酸(Glu)含量的影响及其机制.[方法]利用微量透析法和高效液相色谱-电化学检测法检测侧脑室微量注射给予0.06,0.60 mmol/L NPW30后PVN区Glu含量的变化.[结果]侧脑室微量注射给予0.60 mmol/L NPW30(高剂量组)可引起PVN区Glu水平升高,且伴有动脉血压升高和心率加快,与给药前的基础值、正常对照组和0.06 mmol/L NPW30组(低剂量组)相比较差异均有统计学意义(P<0.05).[结论]中枢性NPW30可能介导PVN内Glu等神经递质参与心血管活动的中枢调节.

【Abstract】 OBJECTIVE To explore the effects and its mechanism of intracerebroventricular microinjection of neuropeptide W30(NPW30) on glutamate content in the hypothalamic paraventricular nucleus(PVN) in conscious rats. METHODS Microdialysis and high-performance liquid chromatography electrochemistry detection technique were used to determine the change of glutamate contents in PVN after intracerebroventricular microinjection of 0.06 and 0.60 mmol/L NPW30. RESULTS Intracerebroventricular microinjection of 0.60 mmol/L NPW30(high dose groups) could increase the levels of glutamate(Glu) in the PVN region, accompanied by increased arterial blood pressure and heart rate, the difference was statistically significant compared with the values in baseline before administration, the control group and 0.06 mmol/L NPW30 group(low dose group), respectively(P<0.05). CONCLUSION Central NPW30 might mediate the central regulation of cardiovascular activities by neurotransmitters such as Glu in the PVN.
