基于SCoT分子标记的月季品种资源遗传多样性分析

【摘要】 采用目标起始密码子多态性(SCoT)技术来分析16个月季品种的遗传多样性.通过对SCoT-PCR反应体系优化和PCR扩增程序探索,从55条SCoT分子标记引物中筛选出9条引物.品种间成对遗传相似度变幅介于0.400 0~0.775 0,9条SCoT标记引物的扩增多态性位点百分率为93.5%,显示月季品种间具有较丰富的遗传多样性.分析UPGMA聚类结果可知:16个月季品种在遗传相似性系数为0.62处被划分成2个聚类分支,第一分支又分为三个亚分支,显示不同月季品种间具有明显的遗传分歧,遗传关系复杂.大花月季、丰花月季与微型月季的亲缘关系相对较近,推测月季品种在驯化进程中存在基因重组和基因渐透现象;藤本月季进化独立,与其他园艺类群月季存在较明显的遗传分歧.

【Abstract】 Genetic diversity of 16 kinds of Rosa hybrid is analyzed by start codon targeted polymorphism(SCoT). Through optimization of SCoT-PCR reaction system and exploration of PCR amplification program, 9 primers are selected from 55 universal SCoT molecular marker primers. The variation of genetic similarity between cultivars ranges from 0.400 0 to 0.775 0, and the percentage of amplified polymorphic loci of 9 SCoT marker primers is 93.5%, which indicats that there is rich genetic diversity among varieties of Rosa hybrid. The results of UPGMA clustering show that the 16 kinds of Rosa hybrid could be divided into two distinct cluster branches at the genetic similarity coefficient of 0.62, and the cluster I could be divided into three distinct sub-branches, indicating that there are obvious genetic differences among different Rosa hybrid varieties, genetic relationships are complex. The genetic relationships among Grandiflora roses, Floribunda Roses and Miniature roses are relatively close, speculating that there occures gene recombination and gene introgression in the domestication process of rose varieties. Rambler roses evolves independently and have obvious genetic differentiation with other horticultural varieties of roses.
