Treg/Th17细胞平衡在反复种植失败患者外周血中的调节作用
Effect of the Treg/Th17 cell balance in peripheral blood of patients with repeated implantation failure
【摘要】 目的探讨Treg/Th17细胞平衡在反复种植失败(recurrent implantation failure,RIF)患者外周血中的调节作用。方法收集来我院行IVF治疗的RIF患者(实验组)和首次移植妊娠成功患者(对照组)外周血及其临床资料各20例。RT-PCR检测两组患者外周血中Treg和Th17特异性转录因子Foxp3、ROR-γt和相关细胞因子IL-10、IL-17的m RNA水平,数据进行统计分析后探讨Treg/Th17调控RIF发生的可能分子机制。结果与对照组相比,实验组Foxp3(0.45±0.03 vs 0.86±0.06)、ROR-γt(1.27±0.75 vs 0.43±0.15)和IL-10(1.03±0.15 vs 1.60±0.08)、IL-17(1.06±0.02 vs 0.57±0.04)的m RNA表达水平均有显著差异,P均<0.05。结论 Treg和Th17特异性转录因子Foxp3、ROR-γt和相关细胞因子IL-10、IL-17的m RNA水平可能参与了RIF患者外周血中Treg/Th17细胞平衡的调节,从而导致RIF的发生发展。
【Abstract】 Objective Explore the regulatory role of Treg/Th17 cell balance in recurrent implantation failure(RIF) patients’ peripheral blood. Methods 20 cases of peripheral blood and clinical data were collected from RIF patients(experimental group)and patients(control group) who underwent IVF treatment in our hospital. Rt-PCR was used to detect the mRNA levels of Treg and th17-specific transcription factors Foxp3,ROR-γt and related cytokines Il-10 and Il-17 in the peripheral blood of the two groups. After statistical analysis,the possible molecular mechanism of Treg/Th17 regulating the occurrence of RIF was discussed.Results Compared with the control group,the mRNA expression levels of Foxp3(0.45 0.03 vs 0.86 0.06),ROR-γt(1.27 0.75 vs0.43 0.15),Il-10(1.03 0.15 vs 1.60 0.08) and Il-17(1.06 0.02 vs 0.57 0.04) were significantly different in the experimental group(P<0.05). Conclusion The mRNA levels of Treg and th17-specific transcription factors Foxp3,ROR-γt and related cytokines Il-10 and Il-17 may be involved in the regulation of Treg/Th17 cell balance in peripheral blood of RIF patients,thus leading to the occurrence and development of RIF.
