Sox2对TGF-α诱导的EMT的调控及其在前列腺癌转移中的作用

【摘要】 目的探究Sox2对TGF-α诱导的EMT的调控及Sox2在前列腺癌转移中的作用。方法利用倒置显微镜观察法检测细胞形态的变化。利用划痕实验、Transwell迁移模型和Transwell Matrigel实验检测细胞迁移和侵袭能力。利用RT-PCR及Western blot技术检测EMT marker分子E-cadherin、Vimentin及转录因子Slug表达。结果 TGF-α可诱导前列腺癌DU145细胞由上皮转为间质细胞表型,促进细胞迁移和侵袭能力;过表达Sox2可增强前列腺癌PC-3细胞迁移和侵袭能力;二者均促进EMT marker分子E-cadherin表达下调、Vimentin表达上调及转录因子Slug表达升高;TGF-α不能诱导低表达Sox2的DU145-Sox2-shRNA2细胞EMT的发生。结论 TGF-α可诱导前列腺癌DU145细胞发生EMT,增强细胞的迁移和侵袭能力,这种作用可能是通过Sox2介导,进而通过其调控转录因子Slug的表达实现的。

【Abstract】 Objective To research the regulation of Sox2 on EMT induced by TGF-α and the role of Sox2 in prostate cancer metastasis. Methods The shape of cells was observed by inverted microscope. Scratch test, Transwell assay and Transwell Matrigel migration assay were used to measure the invasion and migration change. RT-PCR and Western blot assay were used to detect the expression of E-cadherin, Vimentin and Slug. Results The EMT, migration and invasion were induced by TGF-α in DU145 cells. Migration and invasion capacity were increased by overexpression of Sox2 in PC-3 cells. Expression of E-cadherin was markedly decreased, expression of Vimentin and transcription factor Slug were markedly increased by TGF-α or overexpression of Sox2. The EMT was not induced by TGF-α in DU145-Sox2-shRNA2 cells. Conclusion TGF-α could induce prostate cancer cells EMT. Sox2 is involved in TGF-α induced EMT in prostate cancer cells via increasing the expression of Slug.
