PBK/TOPK表达对膀胱癌BIU-87细胞增殖及迁移能力的影响
The influence of PBK/TOPK expression on the proliferation and migration of bladder cancer BIU-87 cells
【摘要】 目的探讨PDZ连接激酶/T-LAK细胞源蛋白激酶(PDZ-binding kinase/T-LAK cell-originated protein kinase,PBK/TOPK)在膀胱癌中的表达及其表达对BIU-87细胞增殖及迁移能力的影响。方法采用实时定量PCR及免疫印迹试验检测40例配对膀胱癌及癌旁正常组织中PBK/TOPK的表达,随后,分别干扰及过表达PBK/TOPK在BIU-87细胞中的表达,检测PBK/TOPK表达对细胞增殖及迁移能力的影响。结果实时定量PCR及免疫印迹试验检测结果一致表明PBK/TOPK在膀胱癌中的表达明显高于癌旁正常组织(P<0. 05)。干扰PBK/TOPK表达后,BIU-87细胞的增殖及迁移能力明显下降(P<0. 05);而PBK/TOPK过表达后,BIU-87细胞的增殖及迁移能力明显增强(P<0. 05)。结论 PBK/TOPK在膀胱癌组织中,其表达有助于膀胱癌细胞的增殖及迁移。
【Abstract】 Objective To investigate the expression of PDZ-binding kinase/T-LAK cell-originated protein kinase( PBK/TOPK) and its influence on the proliferation and migration of BIU-87 bladder cancer cell line. Methods QRT-PCR and Western blot were used to detect PBK/TOPK expression in 40 cases of bladder cancer and matched normal tissues. Then,to evaluate the effect of PBK/TOPK on cell migration and proliferation,PBK/TOPK expression was knocked down and upregulated in BIU-87 cell line respectively. Results QRT-PCR and Western blot demonstrated that PBK/TOPK was highly expressed in bladder cancer tissues compared with those in normal tissues( P<0. 05). Meanwhile,PBK/TOPK knockdown significantly inhibited cell proliferation and migration,but PBK/TOPK overexpression significantly promoted cell proliferation and migration( P <0. 05). Conclusion PBK/TOPK is highly expressed in bladder cancer,which is contributed to cell proliferation and migration.
