精神分裂症病人外周血差异表达miRNA-181b的生物信息学分析

【摘要】 目的:对精神分裂症病人外周血中差异表达的miRNA-181b进行靶基因、转录因子和信号通路的分析。方法:采用DIANA-TarBase数据库分析miRNA-181b靶基因,应用DIANA-miPath V.3软件对miRNA-181b的靶基因进行GO功能富集分析和KEGG信号通路富集分析,运用DIANA-miRGen v3.0数据库分析miRNA-181b的转录因子。结果:miRNA-181b靶基因共1 344个,其中显著表达的共有靶基因是ARHGAP11A、PPA1、YY1和ZC3H12B;miRNA-181b的靶基因广泛参与生物学过程的调节,包括分子功能、RNA结合、细胞组成等过程;KEGG生物信息学分析发现miRNA-181b的靶基因富集了12条信号通路,包括hippo信号通路、p53信号通路、HIF-1信号通路、赖氨酸的降解等;与miRNA-181b显著关联的转录因子有7个,其中JUN、EGR1、MEF2C与精神分裂症有显著关联。结论:对精神分裂症病人外周血中差异表达的miRNA-181b进行生物信息学分析,发现了多个靶基因、信号通路和转录因子与精神分裂症的发病有密切关联。

【Abstract】 Objective:To analyze the target gene,transcription factor and signal pathway of the differential expression of miRNA-181 b in the peripheral blood of schizophrenic patients.Methods:The miRNA-181 b target gene was analyzed using the DIANA-TarBase database,the GO function and KEGG pathway enrichment of miRNA-181 b gene were analyzed using DIANA-miPath V.3 software,and the transcription factors of miRNA-181 b were analyzed using the DIANA-miRGen v3.0 database.Results:There were 1 344 target genes of miRNA-181 b,and the target genes ARHGAP11 A,PPA1,YY1 and ZC3 H12 B significantly expressed.The target genes of miRNA-181 b were widely involved in the regulation of biological processes(including molecular function,RNA binding and cell composition).The results of KEGG bioinformatics analysis showed that the target gene of miRNA-181 b enriched 12 signal pathways,which included the hippo signal pathway,p53 signal pathway,HIF-1 signal pathway and lysine degradation.Seven transcription factors were significantly associated with miRNA-181 b,and JUN,EGR1 and MEF2 C were significantly associated with schizophrenia.Conclusions:The multiple target genes,signal pathways and transcription factors are closely related to the onset of schizophrenia using the bioinformatics analysis of the differential expression of miRNA-181 b in the peripheral blood of schizophrenic patients.
