HIF-1α信号通路参与胎盘内皮增生的可能机制
Possible mechanisms of HIF-1α signaling pathway for participating in placental endothelial hyperplasia
【摘要】 目的探讨缺氧诱导因子-1α(HIF-1α)信号通路参与胎盘内皮增生的分子机制。方法选择不明原因的反复自然流产产妇78例作为观察组,同期选择行人工流产术产妇78例作为对照组,采集两组的胎盘绒毛标本,免疫组化检测HIF-1α和血管内皮生长因子(VEGF)表达水平,同时进行与胎盘微血管密度(MVD)的相关性分析。结果观察组胎盘绒毛组织的VEGF和HIF-1α阳性率都低于对照组(P <0.05),MVD密度显著低于对照组(P <0.01)。在观察组中,MVD密度与VEGF、HIF-1α阳性率呈显著正相关性(P <0.05),VEGF与HIF-1α阳性率呈显著正相关性(P <0.05)。结论胎盘内皮增生是胚胎发育的基础,HIF-1α信号通路可能参与胎盘内皮增生过程,其低表达可能与不明原因复发性流产有关。
【Abstract】 Objective To explore the possible mechanisms of hypoxia-inducible factor-1α(HIF-1α) signaling pathway for participating in placental endothelial hyperplasia. Methods 78 puerperas with unexplained recurrent spontaneous abortion were selected as an observation group, and another 78 puerperas who had chosen to receive artificial abortion operation were selected as a control group. Specimens of placental villi were collected from the two groups. The expression levels of HIF-1α and vascular endothelial growth factor(VEGF) were detected by immuno-histochemistry. Meanwhile, the correlation with placental microvascular density(MVD) was analyzed. Results The positive rate of VEGF and HIF-1α in the placental villus tissue in the observation group was lower than that in the control group(P < 0.05); the density of MVD was significantly lower than that in the control group(P <0.01). In the observation group, there was a significant positive correlation between the density of MVD and the positive rate of VEGF and HIF-1α(P < 0.05). So does that between the positive rate of VEGF and that of HIF-1α(P < 0.05). Conclusion Placental endothelial hyperplasia is the basis of embryonic development. HIF-1α signaling pathway may participate in the process of placental endothelial hyperplasia and its low expression may be correlated to unexplained recurrent spontaneous abortion.
