淫羊藿苷联合冰片对脑缺血-再灌注损伤模型大鼠抗氧化活性、炎症反应和血脑屏障通透性影响
Effects of icariin combined with borneol on antioxidant activity inflammatory reaction and permeability of blood-brain barrier in rats following cerebral ischemic injury
【摘要】 目的:探索淫羊藿苷配伍冰片对大鼠脑缺血-再灌注损伤抗氧化活性、炎症反应和血脑屏障通透性作用及其机制。方法:SD大鼠50只,随机分成假手术组、模型对照组、淫羊藿苷组、冰片组和淫羊藿苷配伍冰片组,每组10只,采用颈总动脉夹闭法制备脑缺血-再灌注损伤模型。缺血0.5h后,再灌注24h分别检测缺血脑组织超氧化物歧化酶(SOD)活性、髓过氧化物酶(MPO)的活性和伊文思蓝(EB)的含量。采用酶联免疫吸附测定(ELISA)法检测脑组织肿瘤坏死因子-α(TNF-α)的含量。结果:淫羊藿苷组、冰片组和淫羊藿苷配伍冰片组大鼠脑组织SOD活性分别为(259.15±1.129)U/mg,(270.96±1.875)U/mg,(301.14±0.248)U/mg;MPO值分别为(0.741±0.229)U/g,(0.692±0.115)U/g,(0.457±0.046)U/g;EB含量分别为(6.375±0.842)μg/g,(5.419±0.160)μg/g,(4.278±0.175)μg/g,与模型对照组比较,淫羊藿苷组、冰片组和淫羊藿苷配伍冰片组差异均有统计学意义,其中以淫羊藿苷配伍冰片组作用最明显(P<0.01)。淫羊藿苷组、冰片组和淫羊藿苷配伍冰片组大鼠脑组织TNF-α含量分别为(0.516±0.134)U/g,(0.712±0.039)U/g,(0.325±0.092)U/g,与模型对照组比较,淫羊藿苷组、冰片组和淫羊藿苷配伍冰片组差异均有统计学意义,其中以淫羊藿苷配伍冰片组作用最明显(P<0.01)。结论:淫羊藿苷配伍冰片对脑缺血-再灌注损伤模型大鼠的脑组织具有神经保护作用。
【Abstract】 Objective:To explore the effects of icariin combined with borneol on superoxide dismutase(SOD)activity,inflammatory reaction and permeability of blood-brain barrier in rats following cerebral ischemia-reperfusion injury and their potential mechanisms.Methods:50 SD rats were divided into sham operation group,mode control group,icariin group,borneol group and icariin combined with borneol groups.Cerebral ischemia-reperfusion model was established with carotid artery clamping method.SOD activity,myeloperoxidase(MPO)activity and the contents of Evans blue(EB)in rat brain were measured at the 24 th hour after the cerebral reperfusion post 0.5 hischemia.Contents of tumor necrosis factor-α(TNF-α)in rat brain were detected by ELISA.Results:The SOD activity in icariin group,borneol group and icariin combined with borneol group were(259.15±1.129)U/mg,(270.96±1.875)U/mg,(301.14±0.248)U/mg respectively.The MPO activity were(0.741±0.229)U/g,(0.692±0.115)U/g,(0.457±0.046)U/g respectively.The EB content were(6.375±0.842)μg/g,(5.419±0.160)μg/g,(4.278±0.175)μg/g respectively.Compared with the model control group,the differences of three factor were satisticall significant,and the effect of icariin combined with borneol group was most obvious(P<0.01).The TNF-αcontent in icariin group,borneol group and icariin combined with borneol group were(0.516±0.134)U/g,(0.712±0.039)U/g,(0.325±0.092)U/g respectively.Compared with the model control group,the differences were satisticall significant,and the effect of icariin combined with borneol group was most obvious(P<0.01).Conclusion:Icariin combined with borneol exerts neuroprotective effects on cerebral ischemia-reperfusion injury in rats.
