【摘要】 目的 :探讨CUG连接蛋白1(CUGBP1)在结直肠癌组织中的表达,以及特异性沉默结直肠癌SW620细胞中CUGBP1基因表达对细胞增殖和侵袭力的影响。方法:选取2015年3月—2017年12月择期行手术治疗的结直肠癌患者107例,免疫组化法检测结直肠癌和癌旁组织中CUGBP1蛋白表达,培养人结直肠癌SW620细胞,分为干扰序列组、阴性对照组和对照组,实时荧光定量PCR技术检测细胞中CUGBP1基因,MTT法检测细胞增殖能力,Transwell小室检测细胞侵袭力。结果:结直肠癌组织中CUGBP1蛋白阳性表达率77.57%,高于癌旁组织的38.32%,差异有统计学意义(χ2=33.826,P=0.000);结直肠癌组织中CUGBP1蛋白表达与分化程度、TNM分期和淋巴结转移有关(P<0.05);干扰序列组细胞中CUGBP1 m RNA相对表达量低于阴性对照组和对照组,差异有统计学意义(P<0.05);干扰序列组细胞24h、48h、72h和96h时吸光度A值低于阴性对照组和对照组,差异有统计学意义(P<0.05);干扰序列组侵袭细胞数少于阴性对照组和对照组,差异有统计学意义(P<0.05)。结论:CUGBP1蛋白在结直肠癌组织中呈高表达,特异性沉默结直肠癌SW620细胞中CUGBP1基因表达可有效抑制细胞增殖和侵袭力。更多还原

【Abstract】 Objective:To investigate the expression of CUG-bingding protein 1(CUGBP1) in colorectal cancer,and the effects of specific silencing of CUGBP1 gene expression on proliferation and invasiveness of colorectal cancer SW620 cells.Methods:A total of 107 cases of patients with colorectal cancer undergoing elective surgery in our hospital were selected from March 2015 to December2017.Immunohistochemical was used to detect the expressions of CUGBP1 proteins in colorectal cancer and adjacent tissues.Human colorectal cancer SW620 cells were cultured and divided into interference sequence group,negative control group and control group.Real-time fluorescence quantitative PCR was used to detect the expression of CUGBP1 gene in cells.MTT assay was used to detect cell proliferation.Transwell chamber was used to detect cell invasiveness.Results:The positive expression rate of CUGBP1 protein in colorectal cancer tissues was 77.57%,which was higher than that of adjacent tissues(38.32%),the difference was statistically significant(χ2=33.826,P =0.000).The expression of CUGBP1 protein in colorectal cancer was related to differentiation,TNM staging and lymph node metastasis(P <0.05).The relative expression of CUGBP1 mRNA in the interference sequence group was lower than that in the negative control group and the control group,the difference was statistically significant(P <0.05).The absorbance A values at 24 h,48 h,72 h and 96 h in the interference sequence group were lower than those in the negative control group and the control group,the difference was statistically significant(P <0.05).The number of invasive cells in the interference sequence group was less than that in the negative control group and the control group,the difference was statistically significant(P <0.05).Conclusion:CUGBP1 protein is highly expressed in colorectal cancer tissues.Specific silencing of CUGBP1 gene expression in colorectal cancer SW620 cells can effectively inhibit cell proliferation and invasiveness.更多还原
