TLR2促进非小细胞肺癌细胞增殖及其作用机制
TLR2 promotes cell proliferation in non-small cell lung cancer and its mechanism
【摘要】 目的研究TLR2(Toll-like receptor 2,TLR2)基因对非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞增殖能力的影响及其对ERK信号通路的调节作用。方法将人非小细胞肺癌细胞株A549细胞分为空白组、空载体组、TLR2基因过表达组,MTT法检测细胞增殖能力,Western blot法检测ERK1/2和p-ERK1/2蛋白表达水平。结果在A549细胞中,过表达TLR2基因可促进非小细胞肺癌细胞增殖并上调ERK1/2磷酸化水平; U0126能抑制过表达TLR2基因对非小细胞肺癌细胞增殖的促进作用。结论TLR2基因可通过调控ERK信号通路促进体外非小细胞肺癌细胞增殖。
【Abstract】 Objective To investigate the regulation of TLR2 on proliferation and ERK pathway in non-small cell lung cancer( NSCLC) cells. Methods A549 cells,the non-small cell lung cancer cell line,were treated with blank,PCMV-myc and PCMV-TLR2,respectively. Then,cell proliferation was test by MTT assay and ERK protein level was tested by Western blot. In addition,the two cell lines were treated with U0126,an ERK1/2 phosphorylation inhibitor.Results Cell proliferation was promoted and the p-ERK1/2 expression was up-regulated by overexpression of TLR2 in A549 cells. Meanwhile,U0126 treatment restrained the promotion of TLR2 on cell proliferation. Conclusion Overexpression of TLR2 promotes cell proliferation through ERK pathway in NSCLC.
