淮河青虾线粒体细胞色素氧化酶Ⅰ序列的研究

【摘要】 目的:研究青虾线粒体细胞色素氧化酶Ⅰ全序列。方法:以青虾肌肉组织为材料,采用苯酚-氯仿法提取青虾DNA,然后对青虾线粒体细胞色素氧化酶Ⅰ基因进行了PCR扩增和测序。结果:青虾线粒体细胞色素氧化酶Ⅰ全序列长度为1535bp,编码蛋白含511个氨基酸残基。其中A、T、C、G分别为30.0%、30.6%、22.5%和16.9%。T+A含量(60.6%)明显高于G+C含量(39.4%)。结论:青虾线粒体细胞色素氧化酶Ⅰ序列的碱基组成与含量与其他沼虾类相似。

【Abstract】 Objective:The complete sequence of mitochondrial cytochrome oxidase Ⅰ gene was analyzed with PCR-sequencing.Methods:Genomic DNA of Macmbrachium nipponensis was extracted by phenolchloroform.The cytochrome oxidase I of Macmbrachium niponensis was amplified by PCR and sequenced.Results:The cytochrome oxidase Ⅰ sequence of Macmbrachium nipponensis was 1535 bp.The encoding protein contains 511 amino acids.The contents of A,T,C and G of cytochrome oxidase Ⅰ sequence were 30.0%,30.6%,22.5%,16.9%,respectively.The content of AT(60.6%)was higher than the content of GC(39.4%).Conclusion:Base composition and contents of cytochrome oxidase Ⅰ in Macmbrachium nipponensis were similar to those in other Macmbrachium.
