亲水性琼脂糖包埋培养的大鼠乳腺癌细胞对人乳腺癌细胞生长影响的体外研究

【摘要】 目的探讨亲水性琼脂糖包埋的大鼠乳腺癌细胞生长上清对人乳腺癌细胞生长的影响。方法应用亲水性琼脂糖包埋大鼠乳腺癌SHZ-88细胞,使其在凝胶内部生长。通过中性红染色显微镜下观察凝胶内生长细胞的活性。收集不同培养时间的上清,作用于人乳腺癌MCF-7细胞,通过CCK-8检测作用不同时间后的细胞活存状况。结果通过中性红染色后显微镜观察,在包埋SHZ-88细胞的凝胶内可见散在的死细胞与成团的活细胞。包埋细胞生长的上清能够抑制人MCF-7细胞生长,包埋细胞生长时间越长获得的上清抑制细胞生长能力越强。结论亲水性琼脂糖包埋的大鼠乳腺癌细胞生长上清能够有效抑制人MCF-7细胞生长。

【Abstract】 Objective To investigate the effect of hydrophilic agarose encapsulated rat breast cancer cellson the growth of human breast cancer cell lines in vitro. Methods The rat breast cancer SHZ-88 cells were encapsulated and cultured in hydrophilic agarose gel. The viability of cells in agrose gel was observed under the microscope by central red staining. The culture supernatant of encapsulated SHZ-88 cells was collected at different culture time. CCK-8 was used to detect the activity of MCF-7 under culture supernatant of encapsulated SHZ-88 cells. Results Neutral red staining showed that scattered dead cells and mass viable cells were observed in the agrose gel of encapsulated SHZ-88 cells. The culture supernatant of the encapsulated SHZ-88 cells inhibited the growth of human MCF-7 cells,and the longer the growth time of the embedded cells,the stronger the inhibition ability of supernatant.Conclusions The proliferation of human breast cancer cell line MCF-7 can be inhibited effectively by hydrophilic agarose encapsulated SHZ-88 rat breast cancer cells.
