血管内皮生长因子在哮喘小鼠的表达及细辛脑的干预作用

【摘要】 目的 (1)观察哮喘小鼠肺组织中血管内皮生长因子(VEGF)的表达变化;(2)研究细辛脑干预对VEGF表达的影响。方法 24只BALB/c雌性小鼠随机分为对照组(A组)、哮喘模型组(B组)、细辛脑干预组(C组),每组各8只。建立小鼠哮喘模型,酶联免疫吸附测定法(ELISA)对各组小鼠支气管肺泡灌洗液(BALF)和血清中VEGF进行定量分析;Western blot检测VEGF表达;免疫组化法检测小鼠肺组织中VEGF的表达。结果 ELISA法测定结果显示:与对照相比,哮喘组的VEGF在血清和BALF中表达量明显升高,差异具有统计学意义(P<0.05);细辛脑干预组的VEGF含量比哮喘组降低(P<0.05)。Western blot检测哮喘组VEGF表达量相比于对照组升高(P<0.05),细辛脑组小鼠VEGF表达量相对于哮喘组降低(P<0.05)。免疫组化结果显示哮喘组大部分肺泡上皮细胞、支气管平滑肌和血管周围VEGF均呈阳性表达,细辛脑组有一定程度的减轻,对照组几乎没有。结论 VEGF在哮喘小鼠的肺组织内高度表达,可能参与气道重塑过程,细辛脑在一定程度上可改善气道重塑的病理生理过程。

【Abstract】 Objective(1) To investigate the changes of vascular endothelial growth factor(VEGF) in asthma mice.(2) To study the regulation role of asarone on expression of VEGF.Methods Twentyfour female BALB/c mice were randomly divided into three groups on average,the control group,the asthma model group,the asarone group.The mice model of asthma was established by the ovalbumin(OVA) challenge methods.VEGF levels in bronchoalveolar lavage fluid(BALF) and serum were measured by enzyme-linked immunosorbent assay(ELISA),the expression of VEGF in the lung was detected by Western blot,the immunohistochemisty was used to evaluate the pathological changes of bronchial and lung tissue and to analyze the expression of VEGF in the lung.Results The ELISA results showed that compared with the group of control there were statistic differences in VEGF content in BLAF and serum level of asthma group,and it was lower in the asarone group than that in the asthma group(P<0.05).The expression of VEGF in the asthma group was higher than that in the control group(P<0.05),the expression of VEGF in the asarone group was lower than that in the asthma group(P<0.05).Immunohistochemical results showed that the VEGF were positive expression in the majority of the alveolar epithelial cells,bronchial smooth muscle and around blood vessels in group B.There was a certain mitigation in group C but none in group A.Conclusions VEGF was highly expressed in the tissue of asthmatic mice and maybe participated in the process of airway remodeling.The asarone can improve the airway remodeling to some extent.
