米非司酮对人宫颈癌裸鼠移植瘤放疗敏感性及其NF-κBp65蛋白表达的影响
Effect of mifepristone on sensitivity of radiotherapy and NF-κBp65 protein expression in transplanted tumor of human cervical cancer in nude mice
【摘要】 目的探讨米非司酮(MIF)对宫颈癌裸鼠移植瘤放疗敏感性的影响及其作用机制。方法建立人宫颈癌裸鼠移植瘤模型,筛选32只荷瘤裸鼠,随机分为对照组、放疗组、MIF组、MIF+放疗组4组,每组8只。定期测肿瘤体积,绘制肿瘤生长曲线;处死裸鼠,取瘤体组织,TUENL法检测肿瘤组织细胞凋亡;Western blotting检测各组NF-κBp65蛋白表达水平并进行定位、半定量分析。结果与其他各组相比,MIF+放疗组的裸鼠肿瘤体积下降最明显(P<0.05),细胞凋亡指数最高,肿瘤组织中NF-κB p65蛋白表达水平较单纯放疗组下降明显,差异具有统计学意义(P<0.05)。结论 MIF可通过抑制NF-κB活性,促进细胞凋亡,增强人宫颈癌裸鼠移植瘤的放疗敏感性。
【Abstract】 Objective To explore the effect of mifepristone on sensitivity of radiotherapy in transplanted tumor of human cervical cancer in nude mice.Methods Transplanted tumor model of human cervical cancer in nude mice was established,32 tumor-bearing nude mice were selected and randomly divided into control group,radiotherapy group,mifepristone group,mifepristone+radiotherapy group,8 mice in each group.The volume of transplanted tumor was detected periodically,and the transplanted tumor growth curve was drawn.The tumor tissues were collected from the sacrificed mice,TUNEL assay was used to detect the apoptosis of tumor tissue;the expressions of NF-κBp65protein in the four groups were detected by Western blotting.Positioning analysis and semi-quantitative analysis were performed.Results Compared with other groups,tumor volume decreased statistically significantly in mifepristone+radiotherapy group(P<0.05);apoptosis index was the highest;the expression levels of NF-κB p65 protein in mifepristone+radiotherapy group was statistically significantly lower than that in radiotherapy group(P<0.05).Conclusion Mifepristone can enhance sensitivity of radiotherapy in transplanted tumor of human cervical cancer in nude mice through inhibiting the activity of NF-κB and promoting cell apoptosis.
