2,4-二叔丁基苯酚为内标高效液相色谱法测定丙泊酚血药浓度

【摘要】 目的建立以2,4-二叔丁基苯酚为内标的高效液相色谱法测定丙泊酚血药浓度方法。方法血清样品用乙腈-甲醇(75:25)混合液蛋白沉淀后取上清液进样分析,以2,4-二叔丁基苯酚为内标,采用UltimateXB-C18色谱柱(4.6 mm×250 mm,5μm),以水(H3PO4调pH=1.8)-乙腈(70∶30)为流动相,流速1.5 mL/min,检测激发波长270 nm。结果该法专属性高,回收率高,日内日间RSD低。丙泊酚血药浓度在0.210.0 mg/L范围内线性关系良好(r2=0.9998),最低检测浓度0.2970 mg/L。结论本方法测定丙泊酚浓度方便、快捷、准确度好、精密度高,为开展血药浓度检测、药动学研究提供方法学依据。

【Abstract】 Objective To establish a detecting method of propofol plasma concentration by HPLC with 2,4-ditert-butyl phenol as internal standard.Methods The supernatant of serum samples were analyzed after protein precipitation by acetonitrile-methyl alcohol(75∶25) mixed liquor.With 2,4-ditert-butyl phenol as internal standard,UltimateXB-C18 chromatographic column(4.6 mm×250 mm,5 μm) was used.Water(H3PO4,pH=1.8)-acetonitrile(70∶30) was used as mobile phase,flow velocity=1.5 mL/min,and detecting excitation wave length=270 nm.Results This method had high specificity and recovery,with low intra-day and inter-day RSD.When the plasma concentration of propofol was between 0.210.0 mg/L,it showed a good linear relation(r2=0.9998),and the lower limit of detection was 0.2970 mg/L.Conclusion This method is convenient and speedy,with high accuracy and precision,providing a methodological basis for plasma concentration detection and pharmacokinetic research.
