流式细胞仪检测氧化亚铁硫杆菌的生长量及活性

【摘要】 以氧化亚铁硫杆菌为研究对象,建立流式细胞仪(FCM)计数细菌和检测细菌活性的方法。研究结果表明,使用FCM能有效计数细菌,其结果与比浊法具有良好的线性关系(R~2>0.9)。细菌被分为5组,按组依次用10%的醋酸处理10min灭活10%、20%、30%、40%和50%的细菌,经Hoechst33342单染色,通过FCM检测,能够获得每组细菌中活菌所占的比例为79.9%、69.9%、61.6%、53.4%和48.1%。5组细菌按组沸水浴灭活处理50%的细菌0s、30s、60s、180s、300s,经Hoechst33342-PI双染色,通过FCM检测,能够获得群体中正常活细菌的比例、完全灭活细菌的比例以及介于两者之间的细菌所占的比例。从而可以获得细菌生存状态的详细信息。

【Abstract】 The bacterial growth and viability was detected by flow cytometry( FCM) using Thiobacillus ferrooxidans( T. ferrooxidans) BY-3 as a representative organism. Compared with turbidimetry,counting bacteria by FCM can be very effective,and a good correlation was observed between these two methods( R~2>0. 9). T. ferrooxidans BY-3 were divided into 5 groups,treated with 10% acetic acid for 10 min,10%,20%,30%,40% and 50% of the bacteria were inactivated in the 5 groups respectively. After Hoechst33342 single-stained,the bacteria viability of each group was detected by FCM( 79. 9%,69. 9%,61. 6%,53. 4% 和 48. 1%). T. ferrooxidans BY-3 were also divided into 5groups,50% bacteria of each group were treated with boiling water for 0 s,30 s,60 s,180 s,300 s.After Hoechst33342/PI double-stained,the bacteria condition of viability was detected by FCM clearly. The normal live bacteria were in 95. 0%,49. 5%,48. 8 %,45. 7% and 42. 1%. The total inactive bacteria were in 3. 3%,29. 7%,30. 4%,31. 2% and 32. 0%. The bacteria which between live and inactive were in 1. 6%,20. 8%,20. 9%,23. 1% and 26%.
