多种金属全冠修复对犬牙龈细胞Ki67表达的影响
Influence on expression of Ki67 in gingival cells of the dogs after restored with three types of full metal crown
【摘要】 目的检测钴铬合金、银钯合金及纯钛金属全冠修复后犬牙龈细胞中Ki67基因mRNA的表达,观察3种金属材料全冠修复对牙龈细胞增殖的影响。方法取5只健康成年雄性实验犬,将每只犬的牙弓分为4个区,以每只犬的尖牙为实验对象,分别行钴铬合金(A组)、银钯合金(B组)及纯钛(C组)金属全冠修复,第4区为空白对照(D组);于修复前及修复后1、2、3个月采集尖牙牙龈组织,用逆转录聚合酶链反应(RT-PCR)法检测牙龈细胞中Ki67基因mRNA的表达。结果修复后1个月,A、B组Ki67基因mRNA的表达均高于D组,差异均有统计学意义(P<0.05);C组Ki67基因mRNA的表达与D组比较,差异无统计学意义(P>0.05)。修复后2、3个月,A、B、C组Ki67基因mRNA的表达与D组比较,差异均无统计学意义(P>0.05)。结论钴铬合金和银钯合金会短暂引起牙龈细胞异常增殖,而纯钛不会导致牙龈细胞异常增殖。
【Abstract】 Objective To observe the expression of Ki67 gene in gingival cells of the dogs after restored with cobaltchrome alloy full crown,palladium-silver alloy full crown and pure titanium full crown,in order to investigate the effects of three clinical common metal materials on proliferation of the gingival cells. Methods Five adult health male experiment dogs were selected,each dog′s dental arch was divided into four sections,and fangs of each dog,which were restored with cobalt-chrome alloy crown(group A),palladium-silver alloy crown(group B) and pure titanium crown(group C) respectively,were taken as experiment while the fourth area was taken as the control group(group D). The gingival tissues of fangs before restoration and 1,2 and 3 months after restoration were collected,and the RT-PCR was adopted to detect the mRNA expression of Ki67 gene in the tissue cells. Results After 1 month restoration,the group A and group B were superior to the group D on the mRNA expression levels of Ki67 gene with statistically significant difference(P<0.05),but the difference on the mRNA expression levels of Ki67 gene between the group C and the group D was not statistically significant(P>0.05). Compared to the group D,the difference of mRNA expression levels of Ki67 gene in the group A,group B and group C after 2 and 3 months restoration had no statistical significance(P>0.05).Conclusion The cobalt-chromium alloy and palladium-silver alloy can induce transient proliferation disorder of gingival cells,while the pure titanium has no obvious effect on the proliferation of gingival cells.
