MB染色检测PKA调节亚基对烟曲霉萌发孢子活性的影响

【摘要】 烟曲霉是一种机会性致病真菌,肺巨噬细胞(AM)通过活性氧物质(ROS)清除萌发的烟曲霉孢子。蛋白激酶A(PKA)参与调节多种生物体内的氧化损伤应答反应。本实验通过亚甲基蓝(MB)染色和再培养试验,检测烟曲霉野生株(WT)与PKA调节亚基(pkaR)缺失株(ΔpkaR)萌发孢子的活性,分析pkaR基因对烟曲霉萌发孢子抗氧化损伤的影响。实验结果表明,ΔpkaR株的萌发孢子,对于氧化压力有更强的抵抗能力,表明pkaR基因参与调节烟曲霉对于氧化损伤的抵抗。

【Abstract】 Aspergillus fumigatus is an opportunistic pathogenic fungus.Alveolar macrophages(AM)kill germinated Aspergillus fumigatus conidia by reactive oxygen species(ROS).Protein kinase A(PKA)takes part in regulating the respond of oxidative damage in many organisms.This research aims at investigating the role of the regulatory subunit of PKA(pkaR)in the oxidative damage resistance of germinated Aspergillus fumigatus conidia by detecting its sensitivity to oxidative damage of a mutant with a defect in pkaR with its parent A.Fumigatus wild type(WT)by methylene blue(MB)staining and recultivation.It was found that germinated conidia ofΔpkaRdisplays stronger oxidative damage resistance than WT.This result implies that the regulatory subunit of PKA of A.Fumigatus plays an important role in regulating its resistance to oxidative damage.
