响应面法优化北沙参中糖类的提取工艺研究
Optimization of Extraction Process of Sucrose from Radix Glehniae by Response Surface Methodology
【摘要】 目的:采用响应曲面法以蔗糖为指标优化北沙参中糖类的提取工艺条件。方法:首先进行单因素试验,分别考察乙醇浓度、提取时间和水料比三因素对蔗糖提取率的影响,在此基础上,进行响应曲面分析,优化北沙参蔗糖的最佳工艺条件。结果:最佳提取工艺条件为乙醇浓度39%、提取时间27 min、水料比57:1,在此条件下,可获得实际平均蔗糖提取率达6.02%。结论:优化工艺稳定可靠,可为中试生产北沙参提供理论依据。
【Abstract】 Objective:To optimize the extracting conditions of carbohydrates of Radix Glehniae with sucrose as the index by response surface methodology.Methods:Firstly,single-factor experiment was conducted,which could investigate the influences of three extraction parameters as ethanol concentration,extraction time and the ratio of water to sample on the extraction yield of sucrose respectively.Secondly,on this basis,RSM was applied to find out the optimum extraction parameter conditions of sucrose from Radix Glehniae.Results:The best experimental conditions were found to be extracted for 27 min using 39% ethanol concentration with an L/S ratio of 57:1.Under this condition,the extraction yield of sucrose from Radix Glehniae at average actually was 6.02%.Conclusion:Optimized extraction technology was stable and reliable,which could provide the theory basis for pilot production of Radix Glehniae.
