模拟失重下空间诱变菌感染巨噬细胞炎症相关microRNAs的筛选及生物信息学分析

【摘要】 目的探索模拟失重下空间诱变大肠杆菌(T1-13)感染巨噬细胞RAW264.7后差异表达的microRNAs(miRNAs)及其意义。方法采用第二代测序技术检测模拟失重染菌组与对照组巨噬细胞miRNAs差异表达,通过生物信息学预测差异miRNAs的靶基因,并对靶基因进行GO功能富集分析、KEGG信号通路分析以及STRING蛋白互作分析。结果模拟失重染菌组与对照组间筛选出33个差异miRNAs,下调23个,上调10个。这些miRNAs互补结合的靶基因参与了MAPK通路、WNT通路、轴突导向以及TGF-β等信号通路,其中关键下调的靶基因是MAPK9,MAPK14,MAP3K7,MAP3K14,PIK3CA,PIK3CB,PIK3CD,MYC,JUN,SMAD4;关键上调的靶基因是AKT3,MAPK8,MAPK9,MAPK10,MAP2K1,PIK3CD。结论模拟失重下空间诱变大肠杆菌感染巨噬细胞后miRNAs表达出现显著差异,这些miRNAs可能通过调控MAPK,WNT以及TGF-β等通路参与失重条件下炎症反应的发生。

【Abstract】 Objective To explore the expression profiles difference and significance of the microRNAs( miRNAs) in RAW264. 7 macrophages infected by E. coli from spaceflight under simulated weightlessness. Methods New generation sequencing was used to detect the different expression of miRNAs in macrophage infected by E. coli T1-13 from spaceflight after simulated weightlessness( MG + T1-13). Bioinformatics method was used to predict the target genes of differentially-expressed miRNAs,the target genes were analyzed with GO enrichment analysis,KEGG pathway analysis and STRING protein interaction analysis. Results Thirty-three miRNAs were screened out from the MG + T1-13 group compared with control,in which 23 genes were downregulated and 10 up-regulated. These target genes complementary binding to miRNAs involved in the MAPK,Wnt and TGF-β signaling pathways,in which down-regulated key target genes are MAPK9,MAPK14,MAP3K7,MAP3K14,PIK3 CA,PIK3CB,PIK3 CD,MYC,JUN and SMAD4,up-regulated key target genes are AKT3,MAPK8,MAPK9,MAPK10,MAP2K1 and PIK3 CD. Conclusion Infected by space mutation E.coli under simulated weightlessness induced changes of miRNAs expression in RAW264. 7 macrophages. These miRNAs may involve in the inflammatory reaction under simulated weightlessness through the regulation of MAPK,Wnt and TGF-β signaling pathway.
