蛇足石杉组培快繁体系的研究

【摘要】 通过对蛇足石杉(Huperzia serrata)的组织培养进行系统研究,建立蛇足石杉的快速繁殖体系。结果表明,100 mg/L AAS+0.5 mg/L孔雀石绿组合对蛇足石杉外植体内生菌的抑制效果较好,外植体存活率为20%。蛇足石杉初代培养的最佳培养基为MS+蔗糖20 g/L+琼脂4.59 g/L+IBA 0.05μg/L+KT 1.4μmol/L+谷氨酰胺0.1 g/L+山农一号0.05%+TDZ 0.3 mg/L,诱导率达80%;继代培养中愈伤组织诱导在MS+1.0 mg/L TDZ+1.0 mg/L 2,4-D的条件下,愈伤组织诱导率可达70%;诱导生根的最佳培养基为1/3MS+1.5 mg/L IBA+2.0 mg/L谷氨酰胺,生根率可达90%。

【Abstract】 This article studied the rapid propagation system for Huperzia serrsta. The results showed that the best combination on killing endogenous bacteria is 100 mg/L AAS+0.5 mg/L Malachite green oxalate,the survival rate of explant is 20%. The best medium for the primary culture of Huperzia serrsta was MS+sucrose 20 g/L+Agar 4.59 g/L+IBA 0.05 μg/L+KT 1.4 μmol/L+glutamine 0.1 g/L+Shannong NO.1 0.05%+TDZ 0.3 mg/L,induction rate was 80%;the Callus germination best medium of subculture for Huperzia serrsta is TDZ 1.0 mg/L+2,4-D 1.0 mg/L. Callus induction rate was 70%;The improved rooting medium for Huperzia serrsta was 1/3 MS+IBA 1.5 mg/L+glutamine 2.0 mg/L,Huperzia serrsta the rooting rate was 90%.
