星点设计-效应面法优化莲心总碱自微乳处方
Formulation optimization of self-microemulsion loaded with total alkaloid from Nelumbinis Plumula by central composite design-response surface method
【摘要】 目的采用星点设计-效应面法优化莲心总碱自微乳处方。方法绘制伪三元相图,确定自微乳化区域。以乳化时间、粒径、载药量和多分散指数为指标,星点设计-效应面法优化处方。结果二次多项式模型拟合效果优于多元线性模型。最佳处方为每100 g自微乳含24.57 g油酸乙酯、22.16 g聚氧乙烯蓖麻油、22.16 g吐温-80、23.70 g PEG-400、7.41 g莲心总碱。结论该方法预测性良好,可用于优化莲心总碱自微乳处方。
【Abstract】 AIM To optimize the formulation of self-microemulsion loaded with total alkaloid from Nelumbinis Plumula by central composite design-response surface method. METHODS The self-microemulsion area was identified through drawn pseudo-ternary phase diagram. With emulsification time,particle size,drug loading and polydispersity index as indices,the formulation was optimized by central composite design-response surface method.RESULTS The fitting effect of second quadratic model was better than that of multiple linear model. The best formulation contained 24. 57 g ethyl oleate,22. 16 g cremophor EL,22. 16 g tween-80,23. 70 g PEG-400 and 7. 41 g total alkaloid from Nelumbinis Plumula for every 100 g self-microemulsion. CONCLUSION With good predictability,this method can be used for the formulation optimization of self-microemulsion loaded with total alkaloid from Nelumbinis Plumula from Nelumbinis Plumula.
